Research Of Corydalis Saxicola Bunting Total Alkaloids On Nude Mouse With Subcutaneous Transplantation Tumor And Bone Metastasis In Lung Cancer A549

Objective: Investigate the effect of CSBTA on nude mouse with subcutaneous transplantation tumor and bone metastasis in lung cancer A549 cell.Methods: 1.The models of the non-small cell lung cancer(A549)xenograft transplantde in the right forelimb of BALB/c nude mouse were established.The tumor-bearing mouse were randomly assigned to four treatment groups: Negative control group(0.2ml/d,ig),CSBTA low-dose group(100mg/kg/d,ig),CSBTA high-dose group(300mg/kg/d,ig),DDP(2mg/kg,ip);Tumor size was measured every other day,and tumor volume was draw tumor growth curve.21 days after the death in mouse weighing tumor weight,calculate tumor inhibition rate;TUNEL detect apoptosis;The protein expression of apoptosis related factors survivin were detected by Western-blot.2.Bone metastasis of lung cancer was established in nude mouse.The nude mouse were randomly assigned to three groups: CSBTA(300mg/kg/d,ig),DDP(2mg/kg,once every other day,ip),NS(0.2ml/d,ig).40 days after X-ray testing;52 days after mice were killed.The bone-specific alkaline phosphatase were detected by BALP ELISA dit.Results: 1.The tumor volumes of Negative control group,CSBTA low-dose group,CSBTA high-dose group,DDP group:(1896±130)mm3,(1467±111)mm3,(1145±113)mm3,(708±62)mm3(P<0.05);The CSBTA group and DDP group transplanted tumor weight was lower than the control group(P<0.05),the inhibited tumor growth rate were 18.38%,29.43%,49.16;2.CSBTA can reduce bone metastases and make bone desruction slow.The test results of BALP: normal nude mouse(1.092±0.214)mg/ml,NS(3.255±1.050)mg/ml,CSBTA(1.963±0.639)mg/ml,DDP(1.385±0.504)mg/ml(P<0.05).Conclusions: CSBTA can inhibit the A549 cell transplantation tumor growth,that maybe a result down-regulation survivin.CSBTA can help to reduce the degree of bone destruction that caused by bone metastasis.
