Abstract/Summary:
ObjectiveCerebral ischemic injury is one of the diseases that affects human health,which has a high morbidity,high morbidity,high mortality characteristics.Electroacupuncture on the regulation of MAPK signal pathway in cerebral ischemia-reperfusion injury has been proved.To explore the possible mechanism of electroacupuncture on JNK gene knockout mice of cerebral ischemia reperfusion injury,JNK knockout mice were used to study the integrality of MAPK signal transduction pathway.Try to provide a scientific basis for electroacupuncture against stroke in clinical treatment.MethodsFifty-four JNK knockout mice were randomly divided into sham operation group,model group and electroacupuncture group.Each group was randomly allocated to three subgroups:2 hours,1 day and 3 days time points.The model of local cerebral ischemia and reperfusion was made by middle cerebral artery occlusion.Electroacupuncture group was treated with acupuncturing with "Chize,Hegu,Zusanli and Sanyinjiao" acupuncture points once a day.Neurological function was evaluated by neurological deficit scores.The expression of p-ERK and p-p38 in the brain of JNK knockout mice were detected by double-labelling immunofluorescence and western blot.And the expression of ERK and p38 mRNA were measured by Q-PCR.Result1.Neurological deficit symptoms:Compared with model group,EA can reduce the neurological deficit score of JNK gene knockout mice in the 3 days time points,which has a statistical significance(p<0.05).2.The IOD of p-ERK and p-p38 in the hippocampus and cortex region:The IOD of p-ERK and p-p38 in the hippocampus and cortex region were similar in the electroacupuncture group.The IOD of p-ERK in the EA group was higher than that in the model group at 1d and 3d after reperfusion(p<0.05),namely the expression of p-ERK in the EA group was higher than that in the model group;while the IOD of p-p38 in EA group was lower than that in model group,namely the expression of p-p38 signal pathway in EA group was lower than that in model group(p<0.05).3.The expression of ERK mRNA and p38 mRNA:Compared with I/R group,the expression of ERK mRNA in EA group was increased at 1d and 3d times,while the expression of p38 mRNA in EA group was significantly decreased at Id and 3d time points(p<0.05).4.The protein expression of p-ERK and p-p38:Compared with the model group,the relative protein expression of p-ERK in JNK gene knockout mice was higher in EA group,while the relative protein expression of p-p38 was lower than that of model group.ConclusionAcupuncturing acupoints of "chize","Hegu","Sanyinjiao" and "Zusanli" can reduce brain injury and alleviate the neurological deficits of cerebral ischemia-reperfusion injury of JNK knockout mice.The mechanism of brain protection may be related to the effect of Electroacupuncture on up-regulation of p-ERK and down-regulation of p-p38 signal pathway in JNK knockout mice simultaneously."Electroacupuncture intervention-Regulation of ERK and p3 8 Signaling Pathway in JNK knockout mice simultaneously-Improvement of neurological function-Inhibition of cerebral ischemia-reperfusion injury",which may the mechanism of electroacupuncture on regulation of cerebral ischemia reperfusion of JNK gene knockout mice.
