| ObjectiveThe main way of neuronal death after cerebral ischemia-reperfusion is cell pyrolysis,and its key regulatory gene is Caspase-1 in the Caspase family.Relevant studies have shown that electroacupuncture can down-regulate cell pyrolysis after cerebral ischemia-reperfusion injury.Caspase-1 is expressed,but its regulatory mechanism is not fully understood.Studies have confirmed that GSDMD is the substrate of Caspase-1 and the executor of pyrolysis.This study took Caspase-1 knockout mice as the research object to observe the effect and mechanism of electroacupuncture on cerebral ischemia-reperfusion injury in Caspase-1 knockout mice and its mechanism.It is a clinical electroacupuncture treatment of ischemic stroke.Provide scientific basis.Methods54 Caspase-1 knockout mice and 54 common wild-type C57BL/6 mice were selected and divided into two groups:Caspase-1 knockout mouse group(KO)and common wild-type C57BL/6 mice Group(WT),each group is randomly divided into 3 subgroups,sham operation group(sham),model group(I/R)and electroacupuncture group(EA),a total of 6 groups,each with 18 rats.In the electroacupuncture treatment group,the four points "Zusanli,Sanyinjiao" and "Chize,Hegu" on the affected side were treated with acupuncture intervention for 20 minutes after successful modeling.Neural function scores were used to evaluate the nerve function of each group of mice,TTC staining method was used to observe the morphology of nerve cell damage,TUNEL to observe the level of ischemic focus cell pyrolysis,Western blot method to detect the expression of Caspase-1 and GSDMD-N protein.Results1.Neurological deficit score:The score of the sham operation group in the genotype group(KO)and wild-type group(WT)is 0,and the rest of the groups all showed different degrees of neurological deficit symptoms(P<0.05).Comparison between genotype group and wild type group:Compared with the model group of the same group,the score of the electroacupuncture group decreased,and the difference was statistically significant(P<0.05).2.TTC staining:pale infarcts of different sizes appeared on the affected side of the brain slices in the 5 groups(P<0.05).Compared with the KO model group,the WT group had a larger infarct size(P<0.05).Comparison within the KO and WT groups:Compared with the model group,the infarct volume of the electroacupuncture group was reduced(P<0.05).3.TUNEL staining:a very small amount of pyrolysis cells were seen in the sham operation group(P>0.05).Compared with the KO model group,there were significantly more pyrolysis cells in the WT group,and the difference was statistically significant(P<0.05).After the intervention of electroacupuncture,the cell pyrolysis index decreased significantly(P<0.05).4.Western Blot detection:Compared with WT,there was no expression of Caspase-1 protein in each group in the KO group(P<0.05),and the expression of GSDMD-N was significantly lower(P<0.05).Comparison within the WT group:Compared with the sham operation group,the protein levels of Caspase-1 and GSDMD-N in the model group increased(P<0.05).After electroacupuncture intervention,the protein expression levels of Caspase-1 and GSDMD-N decreased,and the differen ce was statistically significant(P<0.05).ConclusionsElectroacupuncture of"Hegu,Chize" and "Zusanli,Sanyinjiao" significantly reduced the neurological damage in mice after cerebral ischemia and reperfusion,improved histological results and inhibited pyrolysis,and down-regulated Caspase-1 and GSDMD The protein level expression level suggests that electroacupuncture has a protective effect on cerebral ischemia-reperfusion injury and this effect is at least partially mediated by affecting the expression of Caspase-1;one of its regulatory mechanisms may be through "electroacupuncture stimulation-inhibition of Caspase-1 Expression-inhibition of GSDMD-N-anti pyrolysis-anti-cerebral ischemia-reperfusion injury" to treat cerebrovascular diseases. |
