Flos Daturae is the dry flower of Datura metel L.which belongs to the family of Solananeae.They are distributed in most of our provinces with a long history.It has been recorded in Compendium of Materia Medica and in the 2010 edition of Chinese Pharmacopoeia.To be a traditional Chinese medicine,Flos Daturae can relie-ve cough and asthma,alleviate pain and convulsion,etc.It also has been reported that clinical use of Flos Daturae showed significant effect on psoriasis these years.For the purpose of expanding the resources and finding the material basis of similar or higher pharmacodynamic activity than the flowers,this experiment carried out the study on other parts of Datura metel L.gradually.Daturametel leave is the dry leaves of Datura metel L.which belongs to Datura genus,the family of Solananeae.Modern and practical medicine records that it has effects of alleviating asthma,chills,pain,and be used to treat cough,epilepsy,prolapse and bruises.This article preliminarily studied in the leaves of Datura metel L.,which extracted with EtOH and purified with AB-8 macroperous resin,having some new discovery in the withanolides compounds,and providing the important experimental datas for further exploring the mechanism of its pharmacological activity and clinical applications.In order to conduct a comprehensive rational development,expand medicinal resources,this paper worked on the chemical composition of Daturametel leaves.During our study working on the chemical constituents of the EtOH extract of the leaves of Datura metel L.,30 compounds were isolated with AB-8 macroperous resin,silica.gel,reversed-phase(ODS),Sephadex LH-20 column chromatography and HPLC,of which the structures of 20 compounds were determined according to 'H-NMR,'3C-NMR,HSQC,HMBC,1H-1H COSY and NOESY,combined with the technology of ESI-MS,HR-ESI-MS,CD and so on.Among them,there are 3 withanolide compounds,4 alkaloid compounds,5 flavonoid compounds,5 sesquiterpene compounds and 3 other compounds.In these compounds,there are 3 new withanolide compounds,they are named as daturafolisin A,daturafoliside B,daturafolisin C,and the known compounds named as Anoectochine,L-Tryptophan,naphthisoxazolA,Hyoscine,dihydrovomifolio-O-?-D-glucopyranoside,[(6S,9R)-6-hydroxyl-3-oxo-a-ionol-9-O-?-D-glucopyranoside],debiloside A,citroside A,Staphylionoside D,[quercetin-3-O-2-(E-caffeoyl)-a-Larabinopyranosyl-(1?2)-?-D?glucopyranoside-7-O-?-D-glucopyranoside],Kaempferol-3-O-galactoside-(2?1)-glucoside,Kaepmferol-3,7-di-O-?-D-glucopyranoside,[Kaempferol-7-O-?-D-glucoside],L-Tryptophan,quercetin-7-O-glucoside,congmuyaglyeoside I,[N-butyl-O-a-D-Fructofuranosidase],Tropic acid.7 compoundswere firstly isolated from the Solanaceae.This article primarily studied the chemical constituents of the leaves of Datura metel L.,which will provide a scientific basis for fully utilize and expand a natural plant resources of Datura metel L.,mining the unique and important application value of natural active ingredients and lead compounds,and lay a solid foundation for its evaluating the quality and applying in clinic.Our research has a great significant on the value of academic and broad application prospects.
