Objective:The effects of different concentrations of danbeiyifei recipe on human embryonic lung fibroblasts were observed by in vitro culture.The expression of Notch1 mRNA in human embryonic lung fibroblasts was significantly higher than that in different concentrations,Jagged1 mRNA protein expression to explore the effect of danbeiyifei recipe on pulmonary fibrosis.Methods:The lung fibroblasts were cultured in vitro and treated with different concentrations of danbeiyifei recipe.The patients were divided into?1?control group,?2?danbeiyifei recipe 1×10-3mg/ml group,?3?danbeiyifei recipe 1×10-2mg/ml group?4?danbeiyifei recipe1×10-1mg/ml group?5?danbeiyifei recipe lung side 1 mg / ml group,37?,5%CO2 incubator were cultured 24 h,48h,72 h,microscopic The morphological changes of the cells were observed.MTT method was used to detect the absorbance A of lung fibroblasts at different wavelengths at different concentrations.In vitro culture of lung fibroblasts,with different concentrations of danbeiyifei recipe lung side,divided into?1?control group?2?dexamethasone group?5×10-2mg/ml?group?3?danbeiyifei recipe lung side 1×10-3mg/ml group?4?danbeiyifei recipe lung side 1×10-1mg/ml group.The expression of Notch1 mRNA and Jagged1 mRNA in lung fibroblasts were detected by immunohistochemistry and real-time quantitative PCR.The expression of Notch1 m RNA and Jagged1 m RNA was detected by immunohistochemistry.Result:At the same time,the inhibitory rate of human embryonic lung fibroblast proliferation increased with the increase of Danbeiyifei recipe,and the inhibition rate of Danbeiyifei high recipe group(1×10-1mg/ml)was as same as that of dexamethasone group?P>0.05?.The inhibition rate of Danbeiyifei high concentration group was better than that of low dose group?1×10-3mg/ml??P<0.05?.The inhibition rate of the same recipe of Danbeiyifei was increased with time,and it was the highest at 72 h.The inhibitory rate of proliferation of human embryonic lung fibroblasts was positivelycorrelated with the recipe of Danbeiyifi.Conclusion:Danbeiyifei on human embryonic lung fibroblasts,down-regulated the expression of Notch1 mRNA and Jagged1 mRNA,blocked the conduction of Notch signaling pathway,thereby inhibiting the formation of pulmonary fibrosis,with significant anti-fibrosis.
