| Rhubarb is a perennial herb of Polygonaceae,which is mainly distributed in the temperate and subtropical alpine regions of Asia.In our country,it is mainly distributed in the northwest and southwest regions of Qinghai,Gansu,Sichuan and Yunnan.Rhubarb is bitter in taste and cold-natured with the function of remove accumulation with purgation,heat-clearing and fire-purging、cooling blood and stopping bleeding、dampness-draining anti-icteric and so on.Modern pharmacological studies have shown that rhubarb also has the extensive effects of fighting bacteria,tumour,oxidation and inflammation,protecting liver and gallbladder and so on.In this study,six kinds of different extracts extracted from the rhubarb by using six kinds of different solvents.And the rhubarb extracts were used to bacteriostatic test of 10 kinds of cholecystitis bacteria.It is found that extract of ethanol had the best antibacterial effect.9 compounds were isolated from rhubarb by the way of column chromatography and thin layer chromatography.The compounds with good antibacterial effects were selected by extracorporal antibacteria experiments.In this study,9 kinds of monomeric compounds were isolated and purified by column chromatography and thin layer chromatography,respectively.They were 1: Catechin,2: Emodin,3: Chrysophanol,4:Aloeemodin,5: Physcion,6: 4-(4’-Hydroxyphenyl)-2-butanone-4’-O-(2-O-galloyl-6-O-cinnamoyl)Glucoside(4-(4’-hydroxyphenyl)-2-butanone 4’-O-(2-O-galloyl-6-O-cinnamoyl)glucoside),7: 4-(4’-Hydroxyphenyl)2-butanone 4’-O-(6-O-cinnamoyl)glucoside(4-(4’-hydroxyphenyl)-2-butanone 4’-O-(6-O-cinnamoyl),8:Chrysophanol-1-O-glucoside,9: Chrysophanol-8-O-glucoside(chrysophanol-8-O-glucoside).In this study,10 kinds of common cholecystitis pathogens were selected to carry out extracorporal antibacteria experiments by using the 8monoclonal compounds of rhubarb.The results indicated that compounds2 and 4 had significantly inhibitory effect on the above 10 kinds of bacteria(P <0.01).Compound 1 had bacteriostatic effect on nine kinds of cholecystitis pathogens other than Enterobacter aerogenes.And its bacteriostasis to Staphylococcus aureus was significant(P <0.05).Also it plays a great role of fighting the rest 8 pathogenic bacteria(P <0.01).Compound 3 had remarkable bacteriostatic effect on Klebsiella pneumoniae subspecies(P <0.05).Its antibacterial activity against Proteus,Enterococcus faecalis and Klebsiella(P <0.01)is extremely significant.The inhibitory effect of compound 5 on Proteus bacillus,Streptococcus faecalis,Escherichia coli,Enterobacter aerogenes,Enterococcus faecalis,Klebsiella pneumoniae and Klebsiella pneumoniae pneumonia subspecies was extremely remarkable(P <0.01).The bacteriostatic effect on Bacteria Staphylococcus aureus,Pseudomonas aeruginosa and Enterobacter cloacae is not obvious.The inhibitory effect of compound 6 on Staphylococcus aureus was evident(P <0.05).Its bacteriostasis to Proteus Bacteria,Streptococcus faecalis,Escherichia coli,Enterobacter cloacae,(P <0.01)is extremely apparent.The antibacterial activity of compound 8 on Enterobacteriaceae,Enterobacter aerogenes,Klebsiella pneumoniae pneumonia subspecies and Enterococcus faecium(P <0.01)was significant.The inhibitory effect of compound 9 on Proteus,Enterobacter aerogenes,Enterobacter cloacae,Subspecies,Enterococcus faecalis and Klebsiella pneumoniae were significant(P<0.01).The bacteriostasis of Klebsiella pneumoniae subspecies was distinct(P <0.05).In this study,emodin has the best effect on Staphylococcus aureus,Pseudomonas aeruginosa,Enterobacter aerogenes,Klebsiella pneumoniae and Klebsiella pneumoniae pneumonia subspecies.aloeemodin plays the best role of antibacterial effect of Streptococcus faecalis,Enterobacter cloacae subspecies and Enterococcus faecium.emodin has the best antibacterial effect on Escherichia coli.The compound 6(4-(4’-hydroxyphenyl)-2-butanone 4’-O-(2-O-galloyl-6-O-cinnamoyl)glucoside)has the best antibacterial activity against mycobacterium. |
