At present,the molecular mechanism of secondary spinal cord injury is very complex,and plays an important role in the development of pathological process,inflammation and apoptosis are the most important influencing factors.In this study,we observed the effect of bone marrow mesenchymal stem cell transplantation combined with low dose estrogen on neurological function recovery and inflammatory response after spinal cord injury in rats.Bone marrow mesenchymal stem cells can differentiate into osteoblasts,adipocytes,neuron-like cells,vascular endothelial cells and other cells under specific conditions,is considered to be an ideal target cell for cell replacement therapy and gene therapy in medicine,especially in the field of nervous system injury repair has a good application prospects.Estrogen has been used as an anti-inflammatory,anti-oxidation,inhibition of nerve cell apoptosis in the physiological hormone used in the experiment,avoided the high dose of glucocorticoid shock treatment of spinal cord injury caused by many side effects.In the study,100 male SD rats were randomly divided into 5 groups,15 male Sprague-Dawley(SD)rats only cut the skin without modified Allen,S injury;sixty male S-D rats were randomly divided into operation groups: SCI group,17-? estradiol treatment group,BMSCs traspltation group,BMSCs traspltation combined with 17-?estradiol therapy group.The SCI model was established according to the modified Allen,S contution method.BMSCs were trasplantated into the injured sites of spinal coed immediately after SCI.100 ?g 17-?estradiol was adminisrtated by intramuscular injection on the 1h after SCI.At 1,3,5and7 days after SCI functional outcome measurements were evaluated by the BBB score.The expressions of IL-1??TNF-? in the injured spinal cord were detected by ELISA.The nerve cells apoptosis was counted with TUNEL staining.Experiment result shows:(1)The BBB scores in C?D?E 3 group were elevated gradually compared with B group(P<0.05)after SCI 5th day and after 7th day it the BBB score in group E was higher obviously compared with group C?D(P<0.05).(2)Compared with the group B the level of IL-1??TNF-? in the treatment groups was lower(P<0.05)at the 12h?72h.(3)The rate of TUNEL positive cells in the group E were significantly reduced compared with that in group C and D at 72h(P<0.05).Both low doses of 17-?estradiol and BMSCs can faciliate the recovery of motor function after SCI probably by due to the inhibited oxidative stress.
