| Cervical cancer is one of the most common malignant tumors In the world,second only to the breast malignant tumor,but cervical cancer is can be early prevention and diagnosis of malignan tumors.Cervical Intraepithelial Neoplasia(Cervical Intraepithelial Neoplasia,CIN)as the main disease of Cervical cancer lesion before plays an important role,In order to reduce the incidence and mortality of cervical cancer and improve the prognosis for cancer,clinicians should promptly state before interference and blocking,the lesions completely clear,has important significance for prevention and treatment of cervical cancer.So the biological markers as a kind of specific substances appear gradually in the screening and diagnosis of cervical lesions in the work,particularly important to the prevention and treatment of cervical cancer.P16NK4 A gene is a tumor suppressor gene,that is,the human element cycle dependent kinase(cyclin dependent kinase,CDK)4 inhibitory factor(p16)gene,its expression is a tumor suppressor proteins,in cervical lesions,the expression of p16 increased with the increase of the degree of pathological changes.Human olfactomedin protein 4(olfactomedin4,OLFM4)is a new anti apoptosis protein,can promote the growth of tumor cells,and can accurately reflect the situation of cervical epithelial cell proliferation and differentiation,can well distinguish between normal cervical tissue and cervical intraepithelial neoplasia(CIN),the two kinds of biological markers can well assist diagnosis of cervical lesions,and the grade of cervical lesions progress in prediction,so as to reduce the patients economic expenditure and mental burden.We research object is 2015 to 2016 due to cervical disease in First Affiliated Hospital of Hebei North University from the gynecology clinic,and pathological specimens in gynecological outpatient operation room colposcopy cervical biopsy,the age range was 21-68 years old,the experimental group is colposcopy biopsy pathological diagnosis of cervical intraepithelial neoplasia(CIN),and on the basis of the histological diagnosis results grouped the grade of cervical intraepithelial neoplasia Grade1 50 cases,cervical intraepithelial neoplasia Grade2 50 cases,cervical intraepithelial neoplasia Grade3 50 cases.Colposcopy cervical biopsy were operated by the specialist,30 cases of normal control group was due to uterine benign lesions of uterine specimens.Immunohistochemical method was used to detect the expression of P16 and OLFM4 in the cervical intraepithelial neoplasia(CIN).The positive rates of P16 and OLFM4 in the test group were 88.67% and 87.33%,the positive rates of P16 and OLFM4 in test group were respectively 26.67%and20.00%.The test group were higher than the control group.Therefore,the positive rates of P16 have statistical significance between the test group and the control group(χ2=58.463,P<0.05),the positive rates of OLFM4 have statistical significance(χ2=63.459,P<0.05).P16 in the test group(CINI,CINII,CINIII)positive rates were 76.00%,90.00% and 100.00%,there was significant difference(χ2=112.582,P<0.05),there was statistical differences between each two groups(P<0.05).OLFM4 in the test group(CINI,CINII,CINIII)the positive rates were 76.00%,86.00%and 100.00%,there was significant difference(χ2=124.866,P<0.05),there was statistical differences between each two groups(P<0.05).CINIII group positive rate is highest,CINII group positive rate is second,positive rate is lowest in CINI group.From the above data analysis,there was a significant correlation between the positive rate of P16 in CIN lesions(r=0.613,P<0.05),there was a significant correlation between the positive rate of OLFM4 in CIN lesions(r=0.380,P<0.05),The more serious the CIN condition,the higher P16 and OLFM4 positive rates.The experimental group of P16 and OLFM4 expression correlation analysis showed that the expression of P16 and OLFM4 are correlated,the higher the positive rate of P16,OLFM4 positive rate is high.According to the Spearman rank correlation analysis,there was significant correlation of P16 and OLFM4 co-expression with cervical disease degree(r=0.862,P<0.05).The co-expression rates of P16 and OLFM4 were respectively 0,70.00%,100.00% in the CINII,CINII,CINIII,the higher the co-expression rate of P16 and OLFM4,the higher the degree of cervical lesions.In conclusion,there was significant correlation of P16 and OLFM4 co-expression with cervical disease degree.Moreover,the high expression of P16 and OLFM4 in cervical intraepithelial neoplasia(CIN)showed a positive correlation between the clinical classification,will help improve the cervical intraepithelial neoplasia(CIN)diagnosis consistencily,better diagnosis and assessment of cervical intraepithelial neoplasia(CIN).At the same time,the co-expression rate of P16 and OLFM4 with the severity of cervical lesions increased,the combined detection for triage management of cervical cancer screening,early diagnosis and treatment of diseases,especially can better shunt CINII,providing individualized treatment plan. |
