As a kind of antibacterial drug,antibiotic plays an important role in protection of human health.At the same time,the damage of improper use of antibiotic to human health and ecological environment cannot be underestimated.How to achieve rapid detection of antibiotic to further obtain strict supervision of application amount of antibiotic has become the focus of contemporary researchers.The emergence of quantum dots provides a new platform for quantitative detection of antibiotic.As a new type of nano luminescent material,quantum dots have wide excitation spectrum,adjustable emission wavelength,high fluorescence yield,strong bleaching resistance and spatial compatibility.Therefore,it has been widely used in rapid detection of macromolecules,small molecules and active molecules.Based on fluorescence characteristics of quantum dots,this research detects and analyzes three kinds of antibiotic,metronidazole,mitomycin and sparfloxacin,and establishes three fluorescent biosensors which will be used for rapid detection of metronidazole,mitomycin and sparfloxacin.Main research works in this thesis are as follows:1.A simple fluorescent method for detection of metronidazole based on CdSe/ZnS quantum dotsIn this study,we developed a simple and readily implemented fluorescent method for detection of metronidazole based on the effects of CdSe/ZnS core-shell quantum dots,which can be quenched by metronidazole due to electron transfer.Thus,a detection system for fluorescence quantum dots in metronidazole was developed.Without other inducer,this method can effectively avoid the interference of other co-existed substances with good selection of metronidazole.And was shown here to be fully applicable to the detection of metronidazole in metronidazole injection and urine samples.2.A simple fluorescent method for detection of mitomycin based on CdSe/ZnS quantum dotsBased on fluorescence properties of CdSe/ZnS quantum dots,a new sensitive method for quantitative detection of mitomycin in biologic fluid was developed.mitomycin can be easily combined with weak acid carboxyl on the surface of CdSe/ZnS quantum dots and adsorbed to the surface of quantum dots to form nanocomposite of CdSe/ZnS quantum dots and mitomycin due to weak alkaline amino in the structure,which will further lead to fluorescence quench of quantum dots.Thus,a detection system for fluorescence quantum dots in mitomycin was developed.Without other inducer,this method can effectively avoid the interference of other co-existed substances with good selection of mitomycin.3.A simple fluorescent method for detection of sparfloxacin based on CdSe/ZnS quantum dotsBased on fluorescence properties of CdSe/ZnS quantum dots,a new sensitive method for quantitative detection of sparfloxacin in biologic fluid was developed.Sparfloxacin can be easily combined with weak acid carboxyl on the surface of CdSe/ZnS quantum dots and adsorbed to the surface of quantum dots to form nanocomposite of CdSe/ZnS quantum dots and sparfloxacin due to weak alkaline amino in the structure,which will further lead to fluorescence quench of quantum dots.Thus,a detection system for fluorescence quantum dots in sparfloxacin was developed.Without other inducer,this method can effectively avoid the interference of other co-existed substances with good selection of sparfloxacin.The detection limit of sparfloxacin tested by experiments is 0.027?mol/L.At the same time,this method can be used to rapidly detect sparfloxacin in drug and biological fluid.
