The dried tuberous root of Alisma orientalis(Sam.)Juzep.has been officially listed in Chinese Pharmacopoeia,and mainly cultivated in Fujian,Sichuan and Jiangxi province.Clinical studies showed that this herb can lower plasma cholesterol levels and protect the hepatic system.And triterpenoids are the major chemical constituents of this plant.Firstly,a quantitative determination method of the extract from A.orientalis called colorimetry was established.This method was based on Zimmerman reaction,which was the specific reaction occurring in 3-carbonyl of sterones.The roots of A.orientalis were extracted with alcohol by means of reflux,ultrasound,SFE and PSE methods.The results showed that PSE had higher extraction rate compared to the traditional ones according to the colormetric results and can shorten the extraction time.The optimal condition is determined as T=90?,P=100 bar,t=30 min.With the help of normal-phase silica column and preparative reversed-phase HPLC,twelve compounds was obtained.On the basis of MS and NMR,the structures of compounds were identified as guaia-6,7en-4,1 Odiol(1S,5S,14?,15a)(1),alismoxide(2),16-oxy-11-anhydro-alisol A(3),alisol F(4),alismakectone-B 23-acetate(5),alisol F 24-acetate(6),alisol E 23-acetate(7),25-anhydro alisol F(8),24-acetate alisol A(9),25-anhydro-alisol A(10),11-deoxy-alisol A(11),and 25-ethyl,24-acetate alisol A(12).In order to provide scientific evidence for the stability,storage and usage of the reference substances from A.orientalis,the stability of 24-acetate alisol A in the organic solvents was studied in the research.Last,the method of HPLC-fingerprint analysis was established for the quality control of A.Orientalis,especially for the quality control of triterpenoids in this herb.The 11 batches of A.Orientalis samples were analyzed based on the results of similarity analysis.The method is repeatable and reliable,and proved to be able to effectively control the quality of A.Orientalis.Furthermore,six triterpenoids constituents in samples were indentified by comparing their retention times with those of authentic compounds,which were isolated from the plants.
