| Scholars using a variety of polysaccharides and iron synthesis polysaccharide iron.Looking to give body to provide safe and effective,conducive to the absorption of the new iron supplement.In this thesis,use Omithogalum caudatum Ait dry grass as raw material,extract and purified Omithogalum caudatum Ait polysaccharide(OCAP).Use purified OCAP and ferric chloride under alkaline conditions synthesis OPIC.Systematic study of the optimum conditions for the preparation,physical and chemical properties and structure identification,the main contents are:(1)Water extraction and alcohol precipitation extraction OCAP,use Sevage methods to remove protein,dialysis desalination method to remove inorganic salts and small molecule impurities.(2)The iron content of the OPIC as the main evaluation index,the main factors influence the iron in OPIC are:dosage of solvent,dosage of ferric chloride,the ratio of polysaccharide and dosage of sodium citrate(mass ratio),temperature and pH,do the single factor experiments firstly.Then choose the optimal synthesis method of OPIC.The experiment investigated the influential factors of preparation technology by Plackett-Burman design and optimizing significant factors and level with the central composite design.The optimum conditions for the preparation of the complex were as follows:the dosage of solvent was 70ml,the dosage of ferric chloride was 6mL,the mass ratio was 3.5,the temperature was 80℃ and the pH was 8.(3)OPIC were characterized by many structural analysis methods,such as IR spectroscopy,differential scanning calorimetry,transmission electron microscopy,viscosity and particle size,etc.The results showed that:OPIC stable nature,TEM shows that it’s spherical colloidal particles.Initially confirmed that OPIC is β-FeOOH core linked by oxido and hydroxyl bridges.Thermal gravimetric analysis result shows that OPIC is nature and stability in the range of 27℃-225℃;TEM photo shows that the diameter of OPIC is 40-50 nm,OPIC are spherical and colloidal particles.Moreover there are reunited between the particles;spectral characteristics further confirmed that OPIC is β-FeOOH core linked by oxido and hydroxyl bridges;Particle size and particle size distribution measurement results show that synthesized OPIC’s particle size is about 191.5 nm;The conductance properties of the measurement results show that OCAP accounts for the main role in OPIC in nature of the the conductance,iron core exist in the form of neutral particles.(4)The physical and chemical properties studies have shown that OPIC is not in free form,it formed with OCAP in stable complex.Under conditions of strong acid and heating,polysaccharides are destroyed,trivalent iron freed.OPIC solution have no precipitation occurs within the range of pH 3-12,the solution keep stable,transparent and clarified.The reduction research results show that at 37℃ condition,trivalent iron in OPIC basically been completely reduced into divalent iron about 6 h by ascorbic acid.There is a certain correspondence between OPIC’s trivalent iron dissolution time and its restore time.The viscosity properties measurement results show that OPIC exists not in a linear polymer structure in aqueous solution,but in colloidal form;In the same concentration,the viscosity of OCAP is higher than OPIC,OCAP is a leading role in OPIC in viscosity properties.(5)Experimental results show that OPIC has good solubility properties and restore properties,utilization is high.OPIC is expected to become an effective and synergistic effect of the new blood iron supplement. |
