Ofloxacin is one of the most widely used Fluoroquinolones (FQ), belong to the most promising antibacterial chemotherapeutic agents for the treatment of infections of different etiology and localization in humans and animals. The inhibition of DNA gyrase or topoisomerase ? with FQs leads to the suppression of both gram-positive and gram-negative bacteria.The wide use of Ofloxacin in veterinary medicine poses significant risks to human health associated with the entry of these contaminants into the food chain (via food of animal origin) and pollutions to the environment. The uncontrolled uptake of Ofloxacin into the human body can lead to the development of resistant forms of microorganisms and cause pathological states, such as dysbacteriosis, allergic reactions, suppression of the activity of some enzymes, and so on.Immunochromatographic (ICG) test strip are becoming increasingly common as detection method due to their sensitivity, simplicity, lower cost, shorter time, no need of technical skill to interpret the result and overall it is more affordable than other conventional methods. The ICG strip is used to give visual color indication of the presence or absence of the analyte. However this technology are not easy reproducible by other users due to the complicated procedures in construction.This study with ofloxacin as the research object, colloidal gold as a marker to mark of monoclonal antibody against ofloxacin, according to competitive inhibition immunochromatography principle developed ofloxacin colloidal gold immune chromatography test paper strip and carries on the system optimization. including the optimal conditions for the the binding of colloid gold and antibody , cellulose nitrate film types,coated diluent, the dilution ratio of coated diluent, the dilution ratio of gold labeled antibody,the cellulose nitrate film and gold standard pads.After optimization, the optimum conditions were determined by colloidal gold immunochromatographic test strip and set as: the pH for the binding of colloid gold and antibody was 9, the amount is 18 ?L; the cellulose nitrate film used is MilliporeHF135s, the PB buffer solution was used as coated diluent, PBST was used as the gold conjugate pad treating solution.
