Research purposes:through chemical enhancers of tetrahydropalmatine in corydalis tuber cream and ultrasonic in vitro percutaneous cumulative study on absorption enhancers,chemical enhancers,ultrasonic effect of DL-tetrahydropalmatine of penetration;And study in 0.1W/cm2,0.4W/cm2,0.8W/cm2 ultrasonic power and 300,500,700,900,1100 KHz ultrasonic frequency conditions,accumulation of transdermal absorption of DL-tetrahydropalmatine on ultrasonic frequencies and selective study of power.Research methods: the main take animal experiment method,the experiment used in the chemical and promote infiltration agent of 10%azone + 2%propylene glycol,ultrasonic frequency,300 KHz,500KHz,700 KHz,900KHz,1100 KHz,ultrasonic power for low of 0.1W/cm2,high of0.4W/cm2,0.8W/cm2.The subjects can be divided into without promoting agent without ultrasound group,promoting the permeability separate chemical agent,shortness of single ultrasound group,ultrasound+permeability agent group.In the experiment after 5min,10 min,15min,20 min after extraction respectively receive pool 1 ml of liquid,the 0.45?m organic membrane filtration,placed in the sample bottle,then immediately added 1ml of liquid.Sample bottles of automatic filling,via high phase liquid chromatography(HPLC)analysis,draw relevant conditions peak area of tetrahydropalmatine,compute the accumulation of tetrahydropalmatine uptake.The results of the study:(1)Under the condition of without ultrasonic promote,promote infiltration agent chemical group of tetrahydropalmatine significantly higher mean cumulative absorption without chemical promoting agent group(P <0.05);Under the condition of ultrasonic promoting the permeability,chemical promote infiltration agent group of tetrahydropalmatine significantly higher mean cumulative absorption without chemicalpromoting agent group(P < 0.01).(2)In ultrasonic group alone,300,500,700,500,1100 KHz ultrasonic frequency under the action of tetrahydropalmatine cumulative average absorption promote permeability were significantly higher than without ultrasonic chemical group(P < 0.01);In addition to the 1100 KHz ultrasonic frequency,ultrasonic frequency,300 KHz,500KHz,700 KHz,the average cumulative 900 KHZ ultrasonic frequency absorption promote permeability was also significantly higher than that of single chemical agent group(P <0.01 or P < 0.05).(3)In ultrasonic promote infiltration agent + chemical group,five ultrasonic frequency of tetrahydropalmatine average cumulative uptake were significantly higher than without ultrasonic and promote permeability,no chemical and group(P<0.01);In addition to 900KHz(P=0.127>0.05),the accumulation of the rest of the ultrasonic frequency absorption promote permeability were significantly higher than that of single chemical agent group(P=0.000<0.01).(4)No chemical enhancers,the same ultrasonic frequency,ultrasonic time conditions,ultrasound of low,medium and high power of DL-tetrahydropalmatine in vitro Percutaneous absorption of rollup value does not exist significant differences(P=0.461>0.05);Under the same condition of ultrasonic power,same time,ultrasonic frequency of 300,500,700 KHz of DL-tetrahydropalmatine cumulative uptake mean values were significantly higher than the 900,1100 KHz the cumulative uptake values(P=0.000<0.05).(5)Under joint action of ultrasonic and chemistry to promote infiltration agent,under the condition of same ultrasonic frequency,ultrasonic time,medium,low or high osmotic quantity between the group of ultrasonic power there was no significant difference(P>0.05);Under the same ultrasonic power,ultrasonic time,ultrasonic frequency between five accumulation of osmotic quantity is extremely significant difference(P=0.000<0.01);Ultrasonic frequency and ultrasonic power both interactioneffects on the accumulation of tetrahydropalmatine osmotic quantity is extremely significant(P=0.000<0.01),including 300 KHZ in 0.8W/cm2 high under the condition of ultrasonic power,the highest cumulative osmotic quantity,6.310?g/ml?(6)The same ultrasonic frequency under the condition of 300-1100 KHz,adding chemical to promote infiltration agent of tetrahydropalmatine accumulated uptake were significantly higher than that of without chemical promoting agent accumulated uptake(P<0.05);The same ultrasonic power under the condition of high,medium and low,add chemical to promote infiltration agent of tetrahydropalmatine accumulated uptake were significantly higher than that of without chemical promoting agent accumulated uptake(P<0.05).Research conclusions:(1)Chemical penetration enhancers on traditional Chinese medicine of tetrahydropalmatine in corydalis tuber cream has significant in vitro Percutaneous absorption enhancers.(2)Ultrasound on traditional Chinese medicine of tetrahydropalmatine in corydalis tuber cream has significant in vitro Percutaneous absorption enhancers,and ultrasonic enhancers than chemical enhancers enhancers.(3)Joint use of ultrasonic and chemistry to promote infiltration agent of tetrahydropalmatine has more obvious promoting effect,used in combination with two different way promoting the permeability,does not interfere with each other,but to a greater degree of the promotion of tetrahydropalmatine in vitro percutaneous absorption.(4)A single ultrasonic function under the condition of tetrahydropal matine in vitro transdermal absorption of ultrasonic frequency and power of selective,high power ultrasound(0.8W/cm2),the ultrasonic frequency of300 KHz is to facilitate the best choice for tetrahydropalmatine accumulated uptake increased.(5)Ultrasonic + chemical enhancers under in vitro Percutaneous abso rption of DL-tetrahydropalmatine on the ultrasonic frequency and power areselective,and high power(0.8W/cm2),ultrasonic frequency 300 Khz of ultrasonic power and the ultrasonic frequency interaction is strongest,highest cumulative amount corydalis.(6)Under the action of ultrasonic frequency,ultrasonic power,chemical andpromote the impregnation agent and seepage function to expand,tetrahydropalmatine in vitro percutaneous drug accumulation increased uptake.
