Objective:Ultrasonic transdermal delivery can improve the percuta-neous absorption of drugs. If you want to apply this method to the cl-inical, need to study the optimal ultrasonic conditions of each specific drug. According to the clinical commonly used traditional Chinese me-dicine Rhizoma Corydalis were ultrasonic transdermal experiment study medicine, observation of low intensity ultrasound on tetrahydropalmatine effect of transcutaneous permeation quantity, choose the most suitable for tetrahydropalmatine transdermal ultrasound conditions, to provide the e-xperimental basis for the reasonable selection of powerful highend ultr-asound wave therapy of traditional Chinese analgesic parameters appar-atus and clinical treatment of Rhizoma corydalis.Methods:With high-end ultrasound transdermal therapy instrument of rhizom Corydalis cream in vitro study on the transdermal permeation. Experiment of skin by SD male rat abdomen skin. Observation of0.1W/cm2ultrasonic intensity12ultrasonic frequency (300-1400KHz), the5time (3min,5min,10min,15min,20min) with and without adding chemical enhancement effects of Rhizoma Corydalis cream of tetrahydro-palmatinein cumulative permeation quantity. Measured by HPLC performance liquid chromatograph made in different ultrasonic conditions were the tetrahydr-opalmatine. The data obtained by Excel2003and SPSS18.0statistical s oftware for analysis of variance.Result:1. The tetrahydropalmatine passive diffusion cumulative permeation quantity in the range of15-29μg/ml. Ultrasonic effect of tetrahydropalmatine cumulative permeation quantity in the range of62-185μg/ml.2. Ultrasonic of1000KHz20min, tetrahydropalmatine cumulative per-meation amount for a maximum of184.21μg/ml,6.39times the passive diffusion of20min. Add4%azone and5%propylene glycol, combined with ultrasonic of1000KHz20min, tetrahydropalmatine cumulative per- meation amount for a maximum of506.32μg/ml,17.56times the passive diffusion of20min.3. Analysis of variance showed:ultrasonic frequency, ultrasonic time and chemical enhancers all the three factors of tetrahydropalmatine cumulative permeation quantity has significant effect (P<0.01), chemical enhancers> ultrasonic time>ultrasonic frequency.Conclusion:1. Ultrasound can increase permeability of tetrahydropalmatine in Rhizoma Corydalis, and with the increase of ultrasonic time and increase.2. Ultrasonic frequency of300-1400KHz is suitable for ultrasound of tetrahydropalmatine in Rhizoma Corydalis in transdermal drug delivery, in which1000KHz is the best.3. Chemical enhancers and ultrasonic synergistic effect. In the0.1W/cm2ultrasonic intensity of transdermal drug delivery, must add che-mical e-nhancers of percutaneous penetration amount in order to raise t-he tetra hydropalmatine in Rhizoma Corydalis cream.
