Objective: To explore whether oxycodone pretreatment can prevent myocardial ischemia-reperfusion injury in rats and to investigate the role of PI3K/AKT signaling pathway.Methods: forty-eight male SD rats,weighing 250-320 g,7-8 weeks,were randomly allocated into 4 groups(Each group of 12): sham operation(group S);group I/R;Oxycodone pretreatment group(group OP);LY294002 group(group LY).Myocardial I/R was produced by ligation of the left anterior descending branch of coronary artery for 30 min followed by 120 min reperfusion.At 5 min before ligation via the internal jugular vein injection of oxycodone0.5mg/kg,At 30 min before oxycodone injection,LY294002 1.4 mg/kg was injected intraperitoneal in group LY respectively.,At 5 min before ligation via the internal jugular vein injection of the same dose of saline solution in group S and group I/R.Continuously record of ?lead electrocardiogram(ecg);To determine the activity of the serum creatine kinase isoenzyme(CK-MB)and troponin I(cTn I);By TTC staining method to detect myocardial infarction volume;The left ventricular myocardial tissue were obtained for histopathologic examination(with light microscope)and for determination of the expression of Bax and Bcl-2 in myocardial tissues(by immunohistochemical).Results: Compared with group S,the serum CK-MB,cTnI concentrations were significantly increased,Obvious pathological changes in myocardial tissues,Myocardial infarction area increased,and the expression of Bax and Bcl-2 in myocardial tissues was up-regulated,but the ratio of Bcl-2 / Bax is reduced in I/R group,OP group,LY group.Compared with group I/R,the serum CK-MB,cTn I concentrations were decreased,pathological changes reduced in myocardial tissues,Myocardial infarction area decreased,and the expression of Bcl-2 was up-regulated,Bax was down-regulated,the ratio of Bcl-2 / Bax is increased in myocardial tissues was down-regulated in LY group.Compared with group OP,the serum CK-MB,cTnI concentrations were increased,Obvious pathological changes in myocardial tissues,Myocardial infarction area increased,and the expression of Bcl-2 was down-regulated,Bax was up-regulated,in myocardial tissues was up-regulated,the ratio of Bcl-2 / Bax is reduced in LY group.Conclusion: Oxycodone pretreatment can activate the PI3K/AKT signaling pathway to prevent myocardial ischemia-reperfusion injury in rats.
