Effects Of Sinomenine On Expression Of Bax And Bcl-2 In Skeletal Muscle Ischemia Reperfusion In Rats

Objective: To observe the effect of sinomenine on limb ischemia reperfusion in rats,to explore the effect of sinomenine on the expression of Bcl-2 and Bax in rat skeletal muscle cells,through the establishment of rat limb ischemia reperfusion(ischemia reperfusion I/R)model.Methods: Fifty four male Wistar rats,weighing 180~220 g,were randomly divided into 3groups,each group of 18:sham operation group(group S),ischemia reperfusion injury group(group I/R)and sinomenine group(group SIN).Skeletal muscle ischemia reperfusion injury model was induced by ligating the right hind limb of rats with elastic rubber band for proximal 4h followed by reperfusion in group I/R and SIN.Sinomenine 60 mg.kg-1 was injected intraperitoneally(i.p.)at 30 min before reperfusion in group SIN.The equal volume of normal saline was given instead of sinomenine in groups S and I/R.Six animals in each group were randomly chosen at 0h,4h,24 h after reperfusion(T0~2)to get 3ml of blood to measure serum lactate dehydrogenase(LDH)and creatine kinase(CK).The skeletal muscle specimens were immediately obtained after getting blood,one of the specimens was used to weigh the wet and dry weight,the another was used to detect the expression of Bcl-2 and Bax protein(by immune-histochemistry).Pathological changes in skeletal muscle tissues were observed with light microscopes.Results: The pathological changes were significantly reduced in SIN group as compared with I/R group.Compared with group S,the content of LDH and CK were significantly increased,the expression of Bcl-2 protein were descend,the expression of Bax protein were ascend,while Bcl-2/Bax descend at T0~2 in I/R and SIN groups(P<0.05);Compared with group I/R,the content of LDH and CK were significantly decreased,the expression of Bcl-2 protein were ascend,the expression of Bax protein were descend,while Bcl-2/ Bax ascend at T0~2 in SIN groups(P<0.05).Conclusion: Sinomenine can attenuate limbs ischemia reperfusion injury in rats through regulating the expression of Bcl-2 and Bax,and reducing the apoptosis of skeletal muscle.
