Objective: Tongue squamous cell carcinoma cell line CAL-27 was irradiated by low dose X-ray irradiation to study whether low-dose irradiation induced CAL-27 to produce adaptive response,and to investigate the effect of low dose irradiation of tongue squamous cell carcinoma cell line CAL-27 on the proliferation,cell cycle,apoptosis and the level of reactive oxygen species(ROS)and other biological effects.Methods: We studied the effect of a low-dose pre-irradiation(0.03 Gy – 0.1 Gy)alone or followed by a 2.0 Gy challenging dose 4h later on the proliferation,cell cycle,apoptosis and ROS content of the CAL-27 cell line.CCK-8 was used to detect the ability of cell proliferation inhibition.Cell cycle,apoptosis and ROS were detected by flow cytometry.Results: There was no significant difference in the proliferation inhibition rate between D1 group and control group,D2 group and D1 + D2 group(P>0.05),D2 group and D1 + D2 group compared with D1 group showed significant cell proliferation inhibition(P<0.05);D1 + D2 group,D2 group compared with the control group and D1 group showed significant G2 block after 12 hours,and has not returned to normal after 48 hours(P<0.05);D1 + D2 group and D2 group were significantly higher than those of control group and D1 group on apoptosis(P<0.05),but there was no significant difference between D1 + D2 group and D2 group,control group and D1 group(P>0.05);the levels of ROS in D1 + D2 group and D2 group were significantly higher than those in control group and D1 group(P<0.05),but there was no significant difference between D1 + D2 group and D2 group,control group and D1 group(P>0.05).Conclusion: Received pre-exposure of low inductive dose irradiation then challenged by high doses irradiation could not induced the AR of CAL-27 cells.So it is expected to be pre-irradiated with low doses before radiotherapy to reduce damage to normal tissues and cells and improve the effect of cancer treatment.
