| Objective By observing the changes of T, B, NK cells and BDNF, NGF, TNF, IL-2, IL-6,IL-10 levels before and after aripiprazole treatment in patients with first-episode schizophrenia to provide new ideas for the immune pathogenesis and treatment of first-episode schizophrenia.Methods 65 cases of patients with first-episode schizophrenia were selected as the observation group. The range of age was 29-72, the average age was (41.13±11.02) . 65 patients with first-episode schizophrenia is in line with the international Classification of diseases-10 (international Classification of diseases, the ICD-10) about diagnostic criteria for schizophrenia (F20.0 paranoid, F20.1 hebephrenic, F20.2 tension-type, F20.3 undifferentiated type); Positive and negative symptoms scale (PANSS) total were 260(1-7 scale). All of the patients were first-episode schizophrenia without menstrual period,pregnancy, lactation women and other mental disease and patients with alcohol, tobacco,hobby. 65 cases of patients with first-episode schizophrenia were treated by aripiprazole.The levels of BDNF, NGF, TNF, IL-2, IL-6 and IL-10 in serum and PANSS scores were detected before and after treatment. CD3, CD4, CD8, CD19, CD16 and CD56 levels were tested by Flow cytometry before and after treatment in patients; The mRNA expression of IL-2, TNF, NGF were detected using Real-time quantitative PCR before and after treatment in patients; PANSS scores were compared before and after treatment, and the PANSS score is whether associated with levels of BDNF and NGF in serum.Results After treatment, 18 cases (27.69%) were cured, 27 cases (41.54%) were effective,15 cases (23.08%) were turned for the better, 5 cases (7.69%) were invalid, the total effective rate was 92.31%. The levels of BDNF, NGF, TNF, IL-2 and IL-10 in serum were significantly declined. The change of IL-6 in serun was no significant different. The levels of CD3, CD4, CD8, CD19, CD16 and CD56 were declined. The mRNA expression of IL-2, TNF and NGF were declined. According to Pearson correlation analysis, between serum concentration of BDNF and PANSS common symptom points were a positive correlation (r = 0.434, P = 0.002), between serum concentration of NGF and PANSS negative symptom points were a negative correlation (r = -0.509, P = 0.000).Conclusion This study confirmed that the changes of T, B, NK cells and cytokines from them is closely related to the occurrence of first-episode schizophrenia. The levels of BDNF, NGF, TNF, IL-2 and IL-10 in serum were significantly declined. The levels of CD3, CD4, CD8, CD19, CD16 and CD56 were declined. The mRNA expression of IL-2,TNF and NGF were also declined. The level of BDNF in serum and PANSS scores were positively correlatively, revealed that BDNF can be used as evaluation of general symptoms of schizophrenia patients. The level of NGF in serum and PANSS scores were negatively correlatively, revealed that NGF can be used as a biological indicator of clinical symptoms. |
