Objective: To explore the feasibility of intraperitoneal injection of streptozotocin(STZ)in the establishment of diabetic erectile dysfunction mouse model and the application of electrical stimulation of penile cavernous nerve in mouse model for assessment of penile erectile function.The purpose of the experoment was to investigate whether and how single intracavernous injection of stromal vascular fraction on the recovery of erectile dysfunction in the type1 diabetic mice.Methods: Eight-week-old male C57BL6 J mice were randomly divided into 4 groups: normal control(Normal)group,diabetic mice without treatment(No treatment)group,intracavernous injection of PBS(DM+PBS)group and SVF(DM+SVF)group.The mice of type 1 diabetes mellitus were induced by injected STZ(50 mg / kg)for 5 days continuously.Qualified Diabetic mice were screened for weight and blood glucose levels.20 ul of 1x PBS and freshly isolated SVF(1x105cells / 20ul)of epididymal adipose tissue from e GFP mouse were injected into the corpus cavernosum of PBS group and SVF group respectively after modeling 8 week in diabetic mice which Diabetic Erectile Dysfunction Mice Model was successfully established and confirmed by erectile force measurement.After local injection of 2W,in animal experiments,the ICP was recorded and the MSBP before the electrical stimulation was recorded.ICP / MBSP was used to evaluate the erectile function of mice.The corpus cavernosum tissue was obtained immediately while the mice were sacrificed,and the molecular biology experiment was carried out.The expression levels of VEGF-A,PH3,CD31 and p-e Nos in the corpus cavernosum were detected by immunofluorescence staining.The changes of c GMP concentration in the corpus cavernosum were detected by ELISA.Results: The erectile function of diabetic mice was significantly lower than that of DM group and PBS group(P <0.05).The erectile function of diabetic mice in SVF group was significantly higher than that in DM group and PBS group(P <0.05).SVF increased the expression of VEGF-A in the corpus cavernosum of diabetic mice.SVF induced the proliferation of cavernous vascular endothelial cells in diabetic mice.SVF was induced to induce differentiation of spongiform endothelial cells in diabetic mice.SVF increased the content of vascular endothelial cells in diabetic mice,SVF Increased expression of e Nos phosphorylation in the corpus cavernosum of diabetic mice.SVF increased the concentration of c GMP in the corpus cavernosum of diabetic mice.The expression levels of VEGF-A,PH3,CD31 and p-e Nos in the corpus cavernosum of the SVF group were significantly higher than those in the DM group and the PBS group(P <0.05)?Conclusion: SVF can significantly improve the erectile function of diabetic mice.The mechanism of SVF may beincreases the expression of vascular growth factor in the corpus cavernosum or fuses the host endothelial cells to induce differentiation and proliferation,regenerates the penile vascular endothelial cells,restores the endogenous NO-c GMP pathway and improves erectile function.It provides a certain basis for the support for the clinical treatment of diabetic erectile dysfunction.
