The endometrial carcinoma is a type of malignant tumor commonly occurred in female reproductive systems,whose morbidity is increasing gradually with youthoriented tendency.A large number of researches showed that the endometrial carcinoma is frequently accompanied by obvious changes of mucus barriers,whose main ingredients are mucin.It is a type of large-molecular-weight glycoprotein family modified by advanced O-type glycosylation on the cell surface or secreted by the cell surface.It is also a dense O glycan with tandem repeat sequences.Tn antigen is the core mucin O-glycan,which will form sTn antigens after being modified by sialyl transferase.Tn antigen,modified by various glycosyl transferases,will not be expressed in tissues or cells under normal conditions,but which will show positive reactions once with carbohydrate chain synthesis obstrurtion in the disease occurring process.Therefore,Tn/sTn antigen is a specific marker of abnormal O-type glycosylation reaction.A large number of researchers have verified that mucin's abnormal expression is closely related to occurrence of endometrial carcinoma.However,at present,there are no researches to clarify actions and mechanisms of Oglycan abnormality in the occurrence and development of endometrial carcinoma.This research will conduct systematic exploration into relation as well as clinic correlation of expressions of Tn/sTn antigen in endometrial carcinoma,which will help to know about occurrence and development law of endometrial carcinoma and provide certain theoretical basis for early clinical diagnosis and prognosis.ObjectivesTo analyze relations and difference of expressions of tumor-associated carbohydrate antigen--Tn antigen and sTn antigen in endometrial carcinoma tissues,atypical endometrial tissues and normal endometrial tissues as well as its correlation between clinical pathology and animation prognosis.MethodsThe immumohistochemical staining was adopted to detect expressions of Tn antigen and sTn antigen in endometrial carcinoma tissue microarray,atypical endometrial tissue microarray and normal endometrial tissue microarray.Then,the follow-up visit was conducted on 168 patients with endometrial carcinoma.Later,this paper analyzed expression status of Tn/sTn antigen and then looked for its related correlation by combining patients' clinical and pathological data as well as prognosis.Results1.Tn antigen and sTn antigen were not expressed in normal endometrial tissues,but lowly expressed in atypical endometrial hyperplasia and highly expressed in endometrial carcinoma tissues(P<0.001).2.Tn antigen and sTn antigen are mainly located in cytomembrane and cytoplasm of endometrial carcinoma,which are characterized by coexistence and consistency in the distribution area and on the staining intensity.sTn antigen-positive is basically equal to Tn antigen-positive,which represents abnormal O-type glycosylation reaction.3.Tn/sTn antigen expression showed no correlation between patients' age,pathological stage,lymphatic metastasis,depth of myometrial tumor invasion,vessel carcinoma embolus,cervical involvement(P>0.05).However,Tn antigen showed correlation between tumor classification(P=0.046),and sTn antigen showed correlation between degree of differentiation(P=0.046)and tumor classification(P=0.017).sTn antigen-positive expression rate in middle-differentiated cancer tissues was obviously higher than that of high-differentiated cancer tissues.Further,Tn/sTn positive expression rate in I-type endometrial carcinoma tissues was obviously higher than that of II-type endometrial carcinoma tissues.4.Patients with high Tn/sTn expression were characterized by relatively low survival rate and worse prognosis.It was found by Cox regression model that Tn antigen and sTn antigen can be taken as independent factors to judge bad prognosis of endometrial carcinoma.ConclusionTn/sTn antigen is highly expressed in endometrial carcinoma tissues,both of which are co-existed and consistent and closely related to tumor classification,differentiated degree and patients' prognosis.Meanwhile,Tn/sTn antigens can be taken as independent factors to judge prognosis of patients with endometrial carcinoma.
