Objective: By the isolation and selection of Bifidobacterium in the breast milk of Uygur women from Kashi in Xin Jian,the diversity and phylogenetic development of Bifidobacterium can be identified,and the highly active Bifidobacterium strains can be screened for exploiting nutrition food of the ethnic minorities,regulaing intestinal flora and promoting health.In addition,in order to lay the definite theoretical basis for clarifying the mechanism of the dietary structure of ethnic minorities on gastrointestinal flora and the health-related effects.Methods: In this study,the lactic acid bacteria were isolated from Uygur mothers in Xinjiang Kashi area by modified MRS and BS medium.The genotypes and their genetic relationships of lactic acid bacteria were determined by interspecific PCR and 16 S r RNA gene sequences.(GTG)5-PCR,BOX-PCR and ERIC-PCR were used to further identify the Bifidobacterium strains with high homologous of 16 S r RNA gene were future investigated.Finally,the Bifidobacterium strain,which can inhibit the pathogen,were screened by Oxford Cup method,and the beneficial experiment was carried out.Conclusion:1.A total of 1784 presumptive LAB were isolated from the breast milk of Uygur women in Xin Jiang,and the lactic acid bacteria were divided into Lactobacillus,Bifidobacterium,Lactococcus,Enterococcus and Streptococcus.2.Using the three different primers of(GTG)5,BOXA1 R and ERIC1R/ERIC2 to amplify the different bands and of 23 Bifidobacterium strains by Rep-PCR.Through the analysis and comparison of the three fingerprints,the bacteria belong to Bifidobacterium pseudocatenulatum(8strains),Bifidobacterium longum subsp.infantis(3strains),Bifidobacterium longum subsp.longum(2strains)and Bifidobacterium breve(2strains).3.The Bifidobacterium strain,which can inhibit the pathogen,were screened by Oxford Cup method.The result is the four Bifidobacteria could inhibit all the pathogen,and then it showed that the Bifidobacterium pseudocatenulatum 92 strains could not only be resistant to bile,but also have the ability to grow at p H3.0,and when the bile salt concentration is 1.0%,the growth of the strain can be detected.
