?Objective? To observe the effects of olmesartan and atorvastatin on intimal hyperplasia and the pathway of ACE2/Ang(1-7)/Mas after balloon injury of abdominal aorta in rats,providing a new way in the prevention of restenosis.?Methods? 1.About 60 7-8 weeks old male SD rats were randomly divided into 6 groups and 10 rats in each group: normal control group(Group N),balloon injury control group(Group Sham),balloon injury group(Group BI),balloon injury+olmesartan group(Group O),balloon injury+atorvastatin group(Group A),balloon injury+olmesartan+atorvastatin group(Group Com).2.In addition to Group N and Group Sham,the other groups were operated by abdominal aortas balloon injury.Group Sham was only without balloon injury of abdominal aorta and other operation was the same as surgery groups.3.Group O were gavaged administrated olmesartan and Group A were gavaged administrated atorvastatin daily.Group Com were gavaged both olmesartan and atorvastatin and the the rest of the three groups were gavaged the same volume of physiological saline.4.The rats were sacrificed after 28 days,taking the serous samples which were used for the measurement of Ang(1-7).Some of abdominal aortas were stained with HE to observe morphological changes.Specimens of abdominal aortas were obtained for the following evaluation: intimal area(I),medial area(M),the intimal /medial area ratio.The expression level of ACE2 and Mas receptor in abdominal aorta was detected by Western blot method.?Results? 1.HE staining results : Compared with group N,there was no obvious intima hyperplasia in Group Sham,the intimal was made up of a complete layer of endothelial cells.The intimal area(I)and the intimal /medial area ratio(I/M)of Group Sham were no statistical significance compared with Group N(P<0.05);Compared with Group Sham,there was obvious intima hyperplasia in Group BI,Group O,Group A,and Group Com.The I and I/M of above groups were more than Group Sham(P<0.05);Compared with Group BI,there was less intima hyperplasia in Group O,Group A,and Group Com.The I and I/M of above groups were less than Group BI(P<0.05);The I and I/M of Group O were no statistical significance compared with Group A;Compared with Group O and A,there was less intima hyperplasia in Group Com.The I and I/M of Group Com were less than Group O and A(P<0.05).2.Compared with Group N,there was no Ang(1-7)difference in Group Sham;Compared with Group Sham,there was less Ang(1-7)in Group BI,Group O,Group A,and Group Com(P<0.05);Compared with Group BI,there was more Ang(1-7)in Group O,Group A,and Group Com(P<0.05);.Compared with Group O,there was no Ang(1-7)difference in Group A(P<0.05);Compared with Group O and Group A,there was more Ang(1-7)in Group Com(P<0.05).3.The result of Western Blot: Compared with Group N,the expression of ACE2,Mas proteins was no difference in Group Sham;Compared with Group Sham,lower expression of ACE2,Mas proteins was in Group BI(P<0.05);Compared with Group BI,increased expression of ACE2,Mas proteins was in Group O,Group A,and Group Com(P<0.05);Compared with Group O,the expression of ACE2,Mas proteins was no difference in Group A;Compared with Group O and Group A,increased expression of ACE2,Mas proteins was in Group Com(P<0.05).?Conclusion? 1? Olmesartan and atorvastatin can inhibit restenosis after balloon injury of abdominal aorta in rats,and the combination of olmesartan and atorvastatin to inhibit the action of intima hyperplasia even more significant.It hinted that the combination of olmesartan and atorvastatin have synergy in the prevention of restenosis.2? Olmesartan and atorvastatin may be through an up-regulation in the pathway of ACE2/Ang(1-7)/Mas to prevent restenosis after balloon injury of abdominal aorta in rats...
