Vitamin is a kind of trace organic substance from food which maintenance of normal physiological function.Vitamins play an important role in the human body growth,metabolism,and in the process of development.According to the human body physiological indexes,all sorts of vitamins must reach a certain level that can guarantee the body to maintain normal physiological function.A lack of vitamins will cause corresponding deficiency,so as to affect the human body.At present,many people will long-term use of vitamin drugs as supplement.This not only affects the normal function of the body,but also leads to poisoning.Therefore,we must have the corresponding quality control methods to guarantee the vitamin content of food and medicine.In this study,a novel method was developed for study the retention,isolation and quality control of water-soluble and fat-soluble vitamins with microemulsion liquid chromatography,which provided a new method for determination of vitamins in food and medicine.The main contents of this dissertation are as follows:1.The role of the water-soluble and fat-soluble vitamins(such as pharmacological effects,physiological function,etc.),and the detection methods are introduced briefly.Finally microemulsion liquid chromatography and recent application progresses are reviewed.2.The impacts of following factors on the separation selectivity were investigated:the type of surfactant,the type of oil phase,the type of co-surfactant,pH of mobile phase and column temperature.The optimized and validated method can be used for simultaneous determination of water-soluble vitamins in food and pharmaceuticals.3.At the beginning of this experiment,the water-soluble and fat-soluble vitamins are separated respectively?The impacts of following factors on the separation selectivity wereinvestigated:the type of column,the concentration of surfactant,the type of co-surfactant,the ratio of surfactant to co-surfactant,the type of oil phase,the pH of mobile phase.The retention of water-soluble and fat-soluble vitamins in microemulsion is studied using single factor tests and suitable microemulsion composition is screened to make water-soluble and fat-soluble vitamins achieve baseline separation,respectively.The values are provided by single factor tests to the next experiment(including simultaneous separation of water-soluble and fat-soluble vitamins by gradient microemulsion liquid chromatography).4.Simultaneous separation of water-soluble and fat-soluble vitamins by gradient microemulsion liquid chromatography.Two modes of gradient elute(including microemulsion – water and microemulsion-microemulsion)are investigated.The optimal gradient program can be used to simultaneous separation of water-soluble and fat-soluble vitamins.Methodology is proceeded to verify the accuracy and precision of the method.The proposed method can be applied to simultaneous determination of water-soluble and fat-soluble vitamins vitamins in pharmaceuticals.
