Polysaccharide is important component of all living organisms,also playing an important role in the many basic life process.Chinese medicine polysaccharide is an important part of active ingredients in Chinese medicine and many studies show that polysaccharides have immuno-modulatory,anti-tumor and other functions.Anti-oxidation is crucial for these functions.There is different antioxidant activity for different polysaccharides wHere the compound of polysaccharides from different herbs show better antioxidant activity,show their potentail for further sdudy and development..In order to study the antioxidant activity of the extraction of Lycium barbarum polysaccharides?LBP?,Astragalus polysaccharides?APS?,Jujube polysaccharides?JPS?and the three kinds of polysaccharide compound polysaccharide?PM?.The three kinds of polysaccharides and PM were evaluated and compared by assays of anti-lipid peroxidation free radicals scavenging,antioxidant activities in both of cells.the antioxidant activity of polysaccharides were investigated in liver injury model of mice in vivo.The contents of study were summarized as followed:1.LBP,APS,JPS were extracted by water and alcohol precipitation,Sevag,dialysis to remove small molecule impurities,then freeze-drying.Polysaccharide samples were derivative into monosaccharides by PMP,which were identified with high performance liquid chromatography.The results showed that yield of LBP,APS,and JPS was 1.5 %,1.3 % and 1.1 %.the contents of total sugar was 38.5%,28.4% and 43.3% by phenol sulfuric acid.After high performance liquid chromatography,the results showed that LBP,APS,and JPS were mainly composed of 7 kinds of monosaccharide fucose?mannose,rhamnse,glucose,galactose,xylose,arabinose and fucose?,galacturonic acid and glucuronic acid.2.LBP,APS,JPS and PM were evaluated by scavenging DPPH free radical,superoxide anion free radical,hydroxyl free radical and reducing power detection.LO2cells?human normal liver cells?and HepG2 cells?human liver cancer cells?were damaged by H2O2,then the antioxidant activity of polysaccharides in LO2 and HepG2 cells was detected by MTT assay.The levels of MDA,SOD and GSH-Px were also detected.Results showed that all polysaccharides have the antioxidant activity but lower than VC in vitro.The cell models 5?mol/L H2O2 damage to LO2 cells and 50 ?mol/L H2O2 to HepG2 cells were used.The cell viability was the highest in models of LO2 cell and HepG2 cell when treated with the PM at 80 ?g/mL and 160 ?g/mL,respectively.All polysaccharides reduced MDA content and increased SOD and GSH-Px activities in cells.Four kinds of polysaccharides show antioxidant activity,especially PM at the lower concentration,in cells when damaged by H2O2.3.To study effects of polysaccharides to evaluate their activities to protect against hepatic damage using CCl4-induced acute liver injury model by assay of Alb,AST and ALT.Protective function showed by MDA level,SOD,GSH-Px and CAT activity.Results showed that PML,PMM,PMH,LBP,APS,and JPS could remarkably suppress the increase of the activities of ALT and AST and increase albumin content in serum,and could significantly decrease MDA level,increase SOD,CAT and GSH-Px activity in liver-injured mice induced by CCl4,especially PML showed better.It suggested that polysaccharides could increased the activities of antioxidant enzymes in vivo.PM showed more antioxidant activity than other polysaccharide.
