Dendrobium officinale is a type of long-term epiphytic orchid herbaceous plant that grows in high altitude mountains.It is a traditional precious Chinese medicine which has extremely high medicinal and nourishing effects.In recent years,with the improvement of people's awareness of healthy life,the method for preparing safer D.officinale polysaccharides has been optimized on the basis of the past work.The aqueous two-phase extraction method is the first choice in this experiment because it is safer than traditional methods.In this paper,The optimized aqueous two-phase system is a safe,efficient and cost-saving process.The anti-aging effect of D.officinale in body is determined by measuring the anti-oxidative enzyme activity in aging mice which is feeding with the D.officinale polysaccharides.Compound D.officinale preparations were fed to mice to determine its effects of improving immunity,improving anxiety and depression by organ ratio tests,antioxidative enzyme activity tests,and behavioral tests.Response surface methodology was used to optimize the extraction process of D.officinale polysaccharides by aqueous two-phase extraction.The results showed that the optimal conditions for extracted D.officinale polysaccharides in aqueous two-phase method were as follows:added 3 g PEG6000 and 7 g?NH4?2SO4 in per 20 mL polysaccharides solution to formed a two-phase aqueous system.The extraction rate of D.officinale polysaccharides in this system was 92.1364%.Aqueous two-phase extraction of polysaccharides is a simple,cost-effective process.This process can efficiently remove proteins and get residue-free dna safe polysaccharides.The role of polysaccharides in the enhance of antioxidant capacity of the body was investigated by gavage of D.officinale solution in senile female ICR mice.The body weight,organ weight,organ ratio,activity of SOD,CAT,and MDA in each organ were measured of the mice that were orally administered with polysaccharide solution for 31 days.The results showed that D.officinale could delay body weight loss caused by aging.Organ atrophy and other conditions increased the proportion of organs.All of the SOD and CAT enzyme activities of the liver and spleen of mice in each dose group were increased,and the MDA content was decreased with varying degrees.Compared the indexes of each dose group,the middle dose group was the best.Compared with the control group,the activity of CAT and SOD in the liver increased 25.39%and 46.71%,respectively;the SOD and CAT activities in the spleen increased 32.52%and 128.97%,respectively.antioxidant capacity of compound D.officinale preparation was further measured in vitro and in vivo.The ability to scavenge hydroxy radicals,superoxide anion,and DPPH of D.officinale polysaccharides increased gradually as the concentration of aqueous extracts of compound agent increased.After fed 31 days,naturally aging female ICR mice body weight monitoring,organ weight determination,organ weight,serum and organ enzyme activity tests,mouse weight-bearing swimming experiment,elevated plus-maze lab test were performed.The results showed that the compound preparation did not affect the feeding of the mice,and had little effect on body weight changes.The enzyme activity of the mice that fed with the aqueous extract of the compound formula was better than that of the aging control group.The compound D.officinale can improve the weight-bearing swimming time of the mice,increase the tolerance of the aging body,shorten the mouse's adaptation to the labyrinth,increase the number of individuals entering the open end and prolong their stay time on the basis of not affecting the mobility of the mice,and has a certain anti-anxiety effects.Based on the above experimental results,it is found that D.officinale polysaccharides can enhance the antioxidant capacity of organisms.Compound preparation with D.officinale as the main material can improve the body's anti-aging ability,enhance the body's endurance,improve the cognitive ability of organisms learning,and counteract the anxiety of organisms.This study provides an important theoretical basis for the development of D.officinale health products.
