Objectives To investigate the effects and related mechanism of Sanqing Glucoselowering Compound on apoptosis induced by palmitic acid in murine pancreatic ?-cells in order to provide experimental basis for further application of Sanqing Glucoselowering Compound in preventing and treating diabetes.Methods The apoptosis model of Min6 cell was induced by palmitic acid.The Min6 cells were divided into normal control group,model control group,metformin control group,low concentration of Sanqing Glucose-lowering Compound group,medium concentration of Sanqing Glucose-lowering Compound group and high concentration of Sanqing Glucose-lowering Compound group,then treated these groups with appropriate intervention measures for 24 hours.The morphological changes of cells were observed by the inverted microscope;the nuclear morphological changes of apoptosis were detected with Hoechst33342 staining;the insulin secretion was measured by ELISA;the effects of Sanqing Glucose-lowering Compound on cell activity were detected by CCK-8 kits;the apoptosis rate was detected with Annexin V-FITC&PI;the expression of the caspase 9 was detected by immunocytochemistry;the protein levels of caspase 3 was examined by Western bloot.The experimental data were analyzed by SPSS 17.0.Results Compared with normal control group,palmitic acid can make the cell shrinkage,nucleus fragmentation,decrease the secretion of basal insulin and high glucose stimulated insulin(P<0.05),reduce the cell survival rate of Min6 cell(P<0.05),increase the early apoptosis rate(P<0.05)and the expression of caspase 9 and cleaved caspase 3(P<0.01),reduce the expression of procaspase 3(P<0.05);Sanqing Glucose-lowering Compound with different concentrations and metformin can improve the cell and nuclear morphology,increase the secretion of basal insulin and high glucose stimulated insulin(P<0.05),increase the cell survival rate(P<0.05),reduce the early apoptosis rate(P<0.05)and the expression of caspase 9 and cleaved caspase 3(P<0.05).On the influence of cell viability,the cell survival rate of medium concentration of Sanqing Glucoselowering Compound group is higher than that of the group of Sanqing Glucose-lowering Compound with low and high concentration(P<0.05),the cell survival rate of metformin control group is higher than that of low concentration of Sanqing Glucose-lowering Compound group(P<0.05).On the influence of the level of caspase 3,the effect of Sanqing Glucose-lowering Compound with medium concentration,Sanqing Glucoselowering Compound with high concentration and metformin on reducing the expression of cleaved caspase 3 is better than Sanqing Glucose-lowering Compound with low concentration(P<0.05).There was no statistically significant difference between any two of protective medicine groups in Other changes of the apoptosis in Min6 cell(P>0.05).Conclusions 1 Sanqing Glucose-lowering Compound can regulate the insulin secretion of islet beta cells;2 Sanqing Glucose-lowering Compound can increase the cell activity and inhibit the apoptosis of islet beta cells;3 The protective effect of Sanqing Glucoselowering Compound on islet beta cells may be realized by regulating the expression of caspase 3 and caspase 9.
