Objectives 1 To detect the expression of SST gene in the endometrial carcinoma cell line Ishikawa(well differentiated),HEC-1A(moderately differentiated)and KLE.2 To construct the SST over-expression vector.3 The SST over-expression vector was transfected in an SST low expression cell line,in which the effect of over-expression of SST on cell proliferation,migration and invasion were detected.Methods 1 Ishikawa,HEC-1A and KLE were selected from the endometrial cancer cell lines.The m RNA and protein levels of SST in the three cell lines were detected by real time PCR and western blot.2 Constuction of SST over-expression plasmid and lentiviral vector to verify whether the plasmid and lentiviral vector were successfully constructed by sequencing.SST over-expression plasmid vector was transfected into SST low expression cell line by liposome transfection.SST over-expression lentiviral vector was transfected into SST low expression cell lines after lentivirus packaging.Western blot was used to verify whether SST was over-expressed.3 The Ishikawa cell lines were selected and randomly divided into blank control group(Ish),empty lentiviral group(Ish-ctr)and SST over-expression lentivirus group(Ish-SST).CCK8,scratch test and transwell were used to detect the ability of proliferation,migration and invasion separately.SPSS 19.0 software was used for statistical analysis.4 SPSS 19.0 software was used for statistical analysis.The student's t test was used between the two groups.Multiple-group comparison was using one-way ANOVA,and the comparison of two of them was using the SNK test.Timedependent variables were compared using the variance analysis of repeated measurements,P<0.05 indicates a statistically significant difference.Results 1 Compared with Ishikawa cell line,SST was highly expressed in HEC-1A(P <0.05)and KLE(P <0.05)cell lines.Compared with HEC-1A cells,SST was highly expressed in KLE(P <0.05)cells.Ishikawa cell line was lower expression in m RNA and protein levels.2 SST over-expression vector SST-pc DNA3.1+ and p LVX-SST-m CMVZs Green-PGK-Puro were constructed successfully,and p LVX-SST-m CMV-Zs GreenPGK-Puro was successfully transferred into Ishikawa cell line.3 There was no significant change in the proliferation,migration and invasion of SST over-expressing Ishikawa cells compared with Ishikawa cells in blank control group and empty lentiviral group(P >0.05).Conclusions 1 SST was tend to highly expressed in HEC-1A endometrial cancer cell line.2 Ishikawa cell line that could over-express SST was constructed.3 SST over-expression had no significant effect on the proliferation,migration and invasiveness of endometrial carcinoma cells.
