Immunological Activity And Antitumor Activity Of Huangqibuqitang Extract

Huangqibuqitang comes from Zhong Yi Lin Zheng Bei Yao that written by Qinbowei.The formula is mainly applied to abortion patient owing to debility of thoroughfare and conception channels or deficiency of qi and blood.We have proved that HQBQT had anti-infection function by enhancing the immunity of elegans host in the infection model of caenorhabditis elegans in our previous studies.Based on these results,HQBQT water extract was prepared using Astragalus root,Angelica sinensis and Cinnamomum cassia Presl with mass ratio 10:5:3.Moreover,we determined the ferulic acid and total flavones in the extract respectively according to the determination of symbolic component in Pharmacopoeia of People's Republic of China(2015),thus providing a reference for stability and controllability of the extract quality.Furthermore,effects of HQBQT on immunity were determined according to the detecting items in the Technical Specification for Inspection and Evaluation of Health Products(2003).Female Kunming mice were used to determine the effects on immunity,lavage administration with the dosage of 1.5,3,9 g crude drug/kg weight,negative control(distilled water)and positive control(Xiong Xian Fa Xin),dosing them 30 days continuously.Organs/body weight ratio,cellular immune function,humoral immune function,phagocytosis of monocytes-phagocytes,activity of NK cell were detected,respectively.In addition,we performed an acute toxicity experiment with healthy Kunming mice.The growth inhibition effects of HQBQT extract were determined on several different cancer cells by MTT method.The results were shown as follow:1.The optimized parameters of preparation of HQBQT extract were: decocting two times by water,decocting for 45 minutes with 8 times water the first time,then decocting for 30 minutes with 6 times water the second time.2.Ferulic acid and total flavones were selected as the symbolic component for quality control,which content deter mined by HPLC and ultraviolet spectrophotometry respectively.The results showed that the content of ferulic acid was 167 ?g/g extract and total flavones is 6.18mg/g extract.3.HQBQT extract sample was prepared by maximum solubility of 1.02 extract/mL,and the lavage administration was performed with dosage of 0.4 mL/10 g body weight.The results showed that the Maximum Tolerated Dose(MTD)in mice by oral was more than 40.8 g/kg,which was significantly higher than the toxicologic requirment(?10 g/kg)in the Technical Specification of Health Products.4.HQBQT extract had effects on improving delayed allergy,increasing monocyte-macrophages phagocytosis and activity of NK cells,and had no effect on spleen lymphocyte transforming of mice induced by ConA and the level of serum hemolysin.According to the requirments of enhancing immunity in the Technical Specification for Inspection and Evaluation of Health Products(2003),HQBQT extract has the healthy function of enhancing immunity.5.HQBQT extract was proved to have anti-proliferative activities against A549 lung caner cells,PC12 phaeochromocytoma cells,B16 melanoma cells,HepG2 liver cancer cells,and the inhibition effects were in a time and dose dependent manner.
