Objective: The expression of cyclooxygenase-2(COX-2)and matrix metalloproteinase(14)were measured after the effect of different concentrations of COX-2 inhibitor celecoxib in the ECa109 cell,and to measure the proliferation?invasion and apoptosis of cells simultaneously,discuss the role of COX-2 and MMP-14 in the development and progression of esophageal carcinoma finally?Methods: 1.Human esophageal cancer ECa109 cells were cultured in vitro;2.PANC-1 cells were treated with 0(control group),20(low),60(medium),100(high)?mol / L celecoxib concentrations at different durations(24?48?72h).The proliferation ability the cells were detected by MTT colorimetric assay;The ability of invasion and migration were detected by transwell invasion assay and migration assay;The expression of COX-2 and MMP-14 was detected by enzyme-linked immunosorbent assay(EILISA)Protein expression;3.Statistical analysis of data.Results: 1.The results of ELISA showed that the expression of COX-2 and MMP-14 protein decreased gradually with the concentration of celecoxib in the range of 0-100?mol/L,and the expression of COX-2 and MMP-14 was decreased in a concentration-dependent manner.(P <0.05);The protein expression of COX-2 was 1.581±0.116?1.226±0.089?0.846±0.076 and 0.521±0.082,respectively(P<0.05),the protein expression of MMP-14 was 11.517±0.881?7.905±0.724?5.788±0.750?and 3.663±0.601,respectively(P<0.05).The protein expression of MMP-14 was significant correlated with the protein expression of COX-2 in ECa109 cells(r=0.952,P<0.01).2.MTT assay showed that the proliferation ability of ECa109 cells decreased gradually at 24,48 and 72 h.The proliferation rates of the drugs were 25.52 ± 2.39,40.60 ± 4.06 and 49.42 ± 2.67,respectively,compared with the control group(6.92 ± 1.92)for 24 h after treatment for 20,60 and 100 ?mol / Compared with the control group(7.10 ± 1.74),the proliferation rates of the 20,60,100 ?mol / L group were 43.70 ± 5.20,55.77 ± 4.18 and 66.44 ± 4.03,respectively.After 72 h treatment,the proliferation rate was 7.07 ± 1.22),the proliferation rates of 20,60 and 100 ?mol / L groups were 60.34 ± 4.64,72.09 ± 3.93 and 84.31 ± 3.21,respectively.With the increase of drug concentration,the inhibitory effect was gradually increased,and the inhibitory effect was gradually enhanced with the increase of drug time,The inhibitory effect was time and concentration dependent(P <0.05);3.Transwell invasion assay showed that the number of cells passing through the matrix gel was 300.654 ± 12.558,271.041 ± 10.569,184.003 ± 10.865 and 92.667 ± 7.567 by treated with different concentrations after 24 hours of ECa109 cells(F = 237.182,P <0.05),and in a concentration-dependent manner;4.The results of flow cytometry showed that the apoptosis rate of ECa109 cells increased gradually after treatment with different concentrations of ECa109 cells in a dose-dependent manner;The apoptosis rate of ECa109 cells was 1.700±0.557?13.400±1.735?18.767±1.301 ? 28.300±71.988.(P<0.05);Conclusion: 1.COX-2 inhibitor(celecoxib)attenuated the proliferation of ECa109 cells in a concentrationand time-dependent manner,weakened the invasion and promoted the apoptosis ability of ECa109 cells in a concentration-dependent manner;2.COX-2 inhibitor(celecoxib)may down-regulate the expression of MMP-14 by inhibiting the expression of COX-2,attenuating the proliferation and invasion,promoting apoptosis of ECa109 cells,which play a role in anti-esophageal cancer.
