The Anti-tumor Effect Of Carboplatin,Cisplatin Combinated With Bevacizumab In Human Hepatocellular Carcinoma Xenograft Model In Nude Mice

?Objective? To investigate the anti-tumor effect of Carboplatin,Cisplatin combinated with Bevacizumab in human hepatocellular carcinoma xenograft model in nude mice.?Methods? Construct the human hepatocellular carcinoma cell line Hep G2-R in nude mice liver cancer in situ implantation model,when 42 female nude mice transplanted tumor volume to about 300 mm 3,were randomly divided into 7 groups,6 animals in each group.G1 group: carboplatin group,carboplatin 5mg / kg,iv,continuous 7 days;G3: cisplatin group,cisplatin 4mg / kg,iv,for 7 days;G4 group: bevacizumab + carboplatin group,carboplatin 5mg / kg,iv,for 7 days,the first day of carboplatin to be given 400 ug bevacizumab;G5 group: bevacizumab + cisplatin group,cisplatin group 4mg / kg,iv,for 7 days,the first day of carboplatin to be given 400 ug bevacizumab at the same time;G6 group: combination group 1,carboplatin 5mg / kg at the same time with 400 ug bevacizumab,iv,for 7 days;G7 group: combination group 2,cisplatin 4mg / kg at the same time with 400 ug bevacizumab,iv,for 7 days.During the administration period,the mental state and appetite of the nude mice was observed.The weight of the nude mice and the length of the tumor were measured every other day.Draw the body weight and the tumor volume growth curve.One hour after the end of the treatment,the nude mice were sacrificed,completely strip the tumor tissue,calculate the tumor weight inhibition rate after weight the tumors.?Results? 1.Three BALB / c female nude mice subcutaneously inoculated with Hep G2-R cells 3 to 5 days after tumor formation,tumor formation rate of 100%,the tumor issolid,quality toughness,capsule integrity,visible neovascularization.The situ implantation model on the pancreas of nude mice was successfully constructed.After 53 days of inoculation,the volume of transplanted tumor could be as long as 300 mm 3.During the 7 days of administration,the spirit and appetite of nude mice in chemotherapy group G2,G3,G4,G5,G6,G7 compared with negative control group G1 were more sluggish,slightly worse appetite,reduced toilet volume.2.Compared with the control group,there was no significant difference in body weight between the experimental group and the control group.Compared with the negative control group(G1),carboplatin and cisplatin combined with bevacizumab therapy groups(G4,G5,G6,G7)groupcould significantly inhibit the proliferation of human hepatocellular carcinoma(HCC)in rabbits.At the end of the day the tumor volume of these groups were smaller than the control group,the difference was statistically significant.While carboplatin and cisplatin single drug administration,transplanted tumor volume changes were not statistically significant.3.At the end of treatment,the tumor weight and tumor weight inhibition rate of nude mice were compared: G1 negative control group,G2 carboplatin group,G3 cisplatin group,G4 bevacizumab + carboplatin group,G5 bevacizumab + cisplatin group,G6 combination group 1,G7 combination group 2,the levels of tumor weight and tumor weight inhibition rate were: 0.37±0.09 g;0.27±0.07 g,27.0%;0.32±0.13 g,13.5%;0.18±0.05 g,51.4%;0.13±0.06 g,64.9%;0.22±0.07 g,40.5%;0.18±0.06 g,51.4%.The tumor weight of group G4,G5,G6,G7 were significantly smaller than those in the G1 group(P <0.05).Compared with the control group carboplatin and cisplatin single drug administration group(G2,G3),the liver cancer transplanted tumor have a inhibitory effect,but no statistical difference.According to the tumor weight inhibition rate,the results showed that the inhibitory rate of bevacizumab + cisplatin group(G5)was significantly higher than that of the control group,carboplatin monotherapy group and cisplatin monotherapy group,the difference was statistically significant.In addition,by the difference analysis between groups,we found that the administration of bevacizumab(G4,G5)on day one and the administration of bevacizumab(G6,G7)in seven days,the tumor weight difference was not statistically significant.?Conclusion?Carboplatin or Cisplatin combinated with with different arrangements were significantly inhibited tumor growth,while the group given Bevacizumab for 1 day followed by Cisplatin administration for 7 days had the advantage of efficient and economical.
