Clove stem decoction is a classic prescription,which have a long history of clinical in thethreatment of hiccup,and the effect is significant.The aim of this experiment is to develop the clove stem decoction into modern compound granules for the hiccup based on modern preparation technology.Which provide a reference for rational development and clinical application of Dingxiangshidi Granules.According to the basic theory of Chinese medicine and the physical and chemical properties of effective ingredients in the raw material of the prescription and medicine-using characteristics of Dingxiangshidi Granules.Orthogonal test was adopted to optimize extraction technology of volatile oil with yield of volatile oil as indexe and was used to the inclusion process of volatile oil with the application rate of inclusion compounds and entrapment efficiency of volatile oil as indexes.Taking composite score of the content of gallic acid and the total content of ginsenoside Re,Rb1 and dry extract rate as indexs,orthogonal test was also adopted to optimize water.The results showed that the optimized extraction conditions were: 4 folds of water,distilled 4 h.The optimized inclusion conditions were: the proportion of the volatile oils and ?-CD was1:9,inclusion temperature was 40?,inclusion time was 1 h.The optimal water extraction technology were :12 folds of water per time,decocting for 0.5 h,for 3 times.Moreover,the vacuum concentration(80 ?),spraydrying(outlet air temperature is 80 ?)and pelletizing were taked to prepare the Dingxiangshidi Granules.In the quality study,we tested the solubility,grain size,moisture of the granule according to the requirements of granule in Chinese pharmacopoeia of 2015 edition.At the same time,the extract of this granule was measured.TLC method was used to identify Flos Caryophylli,Zingier Officinale,Panax Ginseng.The TLC identification spots were clear and easily to be recognized,and the negative contrast sample was not disturbed.The HPLC was used to determine the contents of eugenol and ginsenoside Re,ginsenoside Rb1 and study the fingerprinting method,and we found that two methods have good specialization,precision degree and repeatability,and can be used for the quality control of the Dingxiangshidi Granules.In order to verify the relevance of this Dingxiangshidi Granules,the Dingxiangshidi dispensing Granules and the traditional decoction,the content and the transfer rate of eu-genol,ginsenoside Re,Rb1 in this Dingxiangshidi Granules,the Dingxiangshidi dispensing Granules and the traditional decoction were compared,in addition,the fin-gerprint was compared of the three.Compared with the traditional decoction,the transfer rates of eugeno of the Dingxiangshidi Granules and the Dingxiangshidi dispensing Granules ware respectively 3.18 times and 0.29 times that of the traditional decoction,and the transfer rates of the total content of ginsenoside Re,Rb1 of the Dingxiangshidi Granules and the Dingxiangshidi dispensing Granules were respectively 1.23 times and 0.68 times that of the traditional decoction.Moreover,the fingerprint compare results showed that the similarity of the three were all higher than 0.9.
