ObjectiveBy study the effects of different extract methods on the extraction of alkaloid in Chinese Goldthread Rhizome,determin the extraction process;E stablish preparation processes of Qijian Particles.First,study effects of granularity on saponins in Sanchi,Optimization the standard substance used to determin the content of Notoginseng total saponin.Optimization the Determination method of total flavonoids in Euonymus alatus Sieb.Second,Establish processes of extract and purify,molding process and quality standards of Qijian Particles;inspect the influence of temperature and pH on saponins in Sanchi.MethodsThe dissertation includes the following parts:establishment of extraction process,molding process and quality standards of Qijian Particles;The effects of temperature and pH on the stability of three saponins in Sanchi,which are Panax ginseng saponin Rgi,ginsenoside Rbi,and notoginsenoside Ri.The main methods used are:(1)The selection of standard substance:using vanillin-acetic acid colorimetric method,respectively selecting ginsenoside Rbi,Rg1,notoginsenoside Ri and PNS as the standard substance to find the best one to determine the PNS in Sanchi by UV measurement.(2)Effects of granularity on saponins in Sanchi:Using HPLC to determine the content of Panax ginseng saponin Rgi,ginsenoside Rb1,and notoginsenoside Ri of different granularity extraction solution.(3)Optimization of extraction processes:using orthogonal design,choosing the content of total sponin,total flavonoids,Panax ginsenoside Rg1,and ginsenoside Rbi as index,the former two determined by UV spectropHotometry and the latter determined by HPLC.(4)Optimization of molding processes:Choosing absorption rate and particles molding rate as index,select the best excipients and optimize its ratio with extracted powder.(5)Establishment of quality standards:using HPLC,TLC methods to conduct the quantitative and qualitative research and developing the quality standards at last.(6)The effect of temperature and pH on the Panax Notoginsenosidum:set different temperatures,different pH,and using HPLC to determine the content changes of ginsenoside Rgi,ginsenoside Rbi,notoginsenoside Ri in Sanchi on different temperature and pH..Results(1)The selection of standard substance:PNS is selected as the best standard substance to to determine the PNS in Sanchi by UV measurement.(2)Effects of granularity on saponins in Sanchi:a significant concentration difference of Panax ginseng saponin Rgi,ginsenoside Rb1,and notoginsenoside Ri exists in the extracting solution of different granularity.(3)Study on extraction processes:through orthogonal design,the selected best extraction processes was:14 times water,extracting three times,each time for two hours.Best alcohol precipitation process:alcohol concentration of 60%,the relative density of 1.0-1.15(40?),alcohol precipitation time more than 10h.Pilot test conducted three batches.(4)Optimization of molding processes:Dextrin as accessories,using 95%ethanol to pelletize,powdered extract with the same percentage of dextrin.(5)Establishment of quality standards:Establish the determination methods of Panax ginseng saponins Rg1,ginsenoside Rb1,Notoginsenoside Ri using HPLC:octadecylsilane bonded silica as a filler,mobile pHase B acetonitrile-C water,gradient elution(0?12 min 19%B,12?60 min,19%?36%B);detection wavelength 203 nm;flow rate of 1.0 mLˇmin-1,column temperature was 30?.Number of theoretical plates ginsenosides Rgi peak should not be less than 4000.(6)The effect of temperature and pH on the Panax Notoginsenosidum:Experiments shows that temperature and pH have a great impact on the content of three saponins in Sanchi,which are Panax ginseng saponin Rgi,ginsenoside Rbi,and notoginsenoside Ri.During the extraction process,which is rapid heating,ginsenosides Rgi,Rbi,notoginsenoside Ri have undergone a transformation at 100?;Rgi and notoginsenoside Ri transformed more than 70%after heating 12hrs.But the standard substance of the three saponins were relatively stable,almost no conversion occurs during the 12hrs' heating.Conclusion(1)Establish the extraction process and moulding technic by optimisation the extraction conditions,molding conditions;Idetificate the herbal medicines in the prescription with TLC and HPLC,formulate its quality criteria.(2)Studying the effect of temperature and pH value on saponins of Sanchi aqueous extract,the result showed that saponins in the extract is unstable,always has a degradation for a long time heating under 100?,while ginsenoside monomers keep stable,Which shows that temperature is not the only factor that induce the degradation of the saponin in the extract.
