Objective: To investigate the pathologic changes and expression of IL-17/ TGF-?1 caused by the combination of budesonide and montelukast in lung tissue of mice infected with respiratory syncytial virus?RSV?.Methods: 120 BALB/c mice were randomly divided into the blank group,RSV infected group,budesonide treatment group,montelukast treatment group and budesonide combined montelukast treatment group,every group 24 only.The blank group mice were instilled intranasally with 100 ?L 0.9% sodium chloride injection,and the other four groups mice were instilled with 100?l 106 PFU RSV virus solution,then three drug groups were respectively treated with the corresponding drug?nebulized 1mg budesonide and/or gavaged 10mg/kg montelukast?after RSV infection.After the above processing,the mice were sacrificed on the 4th,8th,14 th and 28 th day,and its lung tissues were removed to be respectively observed pathological changes by HE staining and collagen deposition by Masson staining,and to be measured the expression of IL-17 and TGF-?1 by immunohistochemistry.Results: 1.Pulmonary pathological changes showed by HE staining: Compared with the blank group,the obvious inflammatory changes was observed in lung tissue of RSV infected group,which was worse than that in the three drug groups.And the group of the pulmonary inflammatory lesions of budesonide combined montelukast treatment was the worst one among them.2.Collagen deposition in the lung tissue: No obvious blue collagen deposition was found in the blank group.Compared with it,the blue collagen deposition increased in RSV infected group on the 4th,8th and 14 th day?P<0.05?,and there was no significant difference on the 28 th day.Compared with the corresponding time points of RSV infected group,collagen deposition decreased significantly in every drug treatment group on the 4th,8th and 14 th day?P<0.05?.In addition,the collagen deposition of the combination therapy group was lower than that of monotherapy group.3.Expression of IL-17: The expression of IL-17 was weakly positive in the blank group.Compared with it,the expression of IL-17 was increased in RSV infected group on the 4th and 8th day?P<0.05?,while there was no significant difference on the 14 th,28th day.Compared with the corresponding time points of RSV infected group,the expression of IL-17 was decreased in every drug treatment group on the 4th and 8th day?P<0.05?,while there was no obvious difference on the 14 th and 28 th day.The expression of IL-17 in the combination therapy group on the 4th and 8th day was lower than that in monotherapy group?P<0.05?.4.Expression of TGF-?1: The expression of TGF-?1 was weakly positive in the blank group.Compared with it,the expression of TGF-?1 was increased in RSV infected group on the 4th,8th and 14 th day?P<0.05?,but there was no obvious difference on the 28 th day.Compared with the corresponding time points of RSV infected group,the expression of TGF-?1 was decreased in every drug treatment group on the 4th,8th and 14 th day?P<0.05?,while there was no significant difference on the 28 th day.The expression of TGF-?1 in the combination therapy group on the 4th,8th and 14 th day was lower than that in monotherapy group?P<0.05?.Conclusion: 1.There are collagen deposition phenomenon on lung tissue of mice infected by RSV;2.IL-17 and TGF-?1 are involved in the process of RSV-induced inflammatory changes in lung tissue.And TGF-?1 may be associated with RSV-induced collagen deposition;3.In order to reduce the inflammation and collagen deposition of lung tissue caused by RSV,budesonide and montelukast may lower the expression of IL-17 and TGF-?1.4.The combination of budesonide and montelukast in reducing the inflammation and collagen deposition of lung tissue caused by RSV is superior to budesonide alone or montelukast along.
