Objective: To observe the effects of Ping-wei capsule on the expression of substance P(SP),vasoactive intestinal peptide(VIP)and calcitonin gene related peptide(CGRP)in the rats of live stagnation and spleen deficiency with functional dyspepsia(FD),using brain-gut axis to explore its mechanism in FD therapy.Methods: 90 Wistar male rats were randomly divided into healthy group and model group.The control group rats were normally fed,but the model rats were established by multiple factors of clasping tails,overstrain and irregular feeding.After 10 days of modeling,every 6 rats in control and model group were randomly selected.Then we observed pathological changes in gastric tissues after gastric emptying,intestinal propulsion and HE staining,to testify whether the modeling was successful.65 rats were randomly divided into 5 groups after modeling: Group FD model,Group with domperidone,Group with high doses of Ping-wei capsule,Group with medium doses of Ping-wei capsule,Group with low doses of Ping-wei capsule,each of 13 rats.The dose given by gastrotube of administration were Ping-wei capsule suspension at 0.24g/ml,0.16g/ml,0.08g/ml for high,medium and low doses of Ping-wei capsule groups,motilium suspension at 0.5mg/ml for group with domperidone,physiological saline for healthy group and FD model group.Each according to 1ml/100 g by gavage twice a day for 3 weeks.All rats were killed and after 3 weeks of administration.Then,gastric evacuation and intestinal propelling were performed.During the experiment,the general condition,body weight and food intake of the rats were observed and recorded,and the pathological changes of gastric tissue were observed under light micorscope.the levels of substance P(SP),vasoactive intestinal peptide(VIP),calcitonin gene-related peptide(CGRP)were detected in serum by enzyme-linked immunosorbent assay(ELISA),levels of SP and VIP in antrum duodenum and hypothalamus were detected by reverse transcription –polymerase chain reaction(RT-PCR)and immunohistochemistry S-P method.Results: 1 Observation of general condition Rats of model group were apathy to stimulation,dysphoria occasionally,unstable mood and loose stool compared with normal group,while groups with administration were high sensitive and active,high sense of mental condition,stool well formed and moderate desiccated compared with model group.2 Food intake and body weight Food intake and weight were decreased significantly in model group rats compared to that of healthy group,and the difference was significant(P<0.01).Weight and food intake were increased significantly in drug-treated group(P<0.01,P<0.05)compared with model group.3 Gastric emptying and intestinal propulsion Rates of gastric emptying and intestinal propulsion were decreased significantly in model group compared to that of healthy group(P<0.01),Rates of gastric emptying and intestinal propulsion were increased significantly of group with medicines(P<0.01,P<0.05)compared with model group.4 Histopathological changes Light microscopic observation on gastric tissues in different groups of rats showed the structure array of mucosal epithelium of each layer being integritly,and little inflammatory cell infiltration were observed in model group.5 Levels amd expressions of serum SP,VIP and CGRRP in rats Plasma SP level was decreased significantly while CGRP and VIP levels were increased significantly in the model group compared with normal group(P<0.01),SPand VIP protein and gene expression increased significantly in the antrum,duodenum and hypothalamus compared with the normal group(P<0.01,P<0.05).Campared with the model group,the plasma CGRP and VIP proteins levels were decreased significantly and SP level increased significantly in groups with medicine(P<0.01,P<0.05),SP and VIP protein and m RNA were decreased significantly in the antrum,duodenum and hypothalamus compared with the normal group(P<0.05,P<0.01).Conclusion: 1 Rats with functional dyspepsia(FD)of depression of live-energy and asthenia of spleen can be established by multiple factors of clasping tails,overstrain and irregular feeding.2 Plasma SP protein level was increased and the plasma CGRP and VIP levels were decreased,SP and VIP protein and m RNA decreased in the antrum,duodenum and hypothalamus with Pingwei capsule,which indicate the relationship between the cause of FD and changing levels of SP,VIP and CGRP.3 In FD therapy of depression of live-energy and asthenia of spleen with Pingwei capsule,using brain-gut axis to correct abnormal expression and emission of brain-gut peptide may be its mechanism,through restoring dynamic balance between brain and gut,improving gastrointestinal motility and visceral hypersensitivity in order to take effect.
