| In this paper,the extraction and isolation of solanesol from potato stems and leaves were studied,and the pre-treatment of potato stems and leaves were studied in order to separate the raw materials of solanil,which was an important medicinal raw material for potato stems and leaves.The research on the saponification process of solanesol and the variation of the solanesol content in different varieties of potato during the growing season were studied.The main research contents included the following aspects:1.Study on extraction technology of solanesol.The extraction efficiency of solanesol was studied by three different extraction methods,such as extraction of cold leaching,extraction of heating and extraction,and extraction of methanol and different concentrations of ethanol as extraction solvent.And the parameters such as the number of extraction and the extraction time are optimized.The optimum conditions were as follows: 95% ethanol was heated and refluxed twice for 1.5 h each time.Under the same conditions,the extraction rate of solanesol was 96.86%.Meanwhile,in order to reduce the cost,the effects of different concentrations of ethanol on the extraction rate of solanesol were investigated.The results showed that after the ethanol concentration reached 80% Little change,so in industrial production can be used as 80% ethanol extraction solvent.2.Study on pretreatment of potato stems and leaves.The effects of enzymatic hydrolysis and high-speed shearing techniques on the extraction efficiency of solanesol were studied.The results showed that the extraction rate of solanesol was 91.38%,which was much higher than that of 79.98% without enzymolysis,and the yield of the extract was only 8.02 %,Significantly lower than 23.29% when not enzymatic hydrolysis.3.Study on saponification of solanesyl ester in this paper,the saponification of solanesol saponification into solanesol was studied by using the concentrate of potato stems and leaves extract as the research object.The optimum saponification conditions were determined by optimizing the conditions such as saponification reagent,saponification temperature,saponification temperature and saponification time.NaOH was used as saponification reagent,the concentration of sodium hydroxide 0.75 mol·L-1,saponification time 0.5 h,saponification temperature 40°C.under the optimized conditions,the quality of solanesol in the extract concentrate increased by 77.82%.4.Simultaneous Extraction and Saponification of Solanesol from Potato Stem and Leaf.Simultaneous extraction of saponified potato stem and leaf solanes was carried out by adding 0.75 mol L·L-1 NaOH with 80% ethanol and petroleum ether as extraction solvent.The results showed that the simultaneous extraction of petroleum ether and the effect of saponification of solanesol were better than 80% ethanol,and the yield of the extract was slightly lower than that of 80% ethanol.However,due to the low boiling point of petroleum ether and long boiling range,Under the conditions of equipment can be used for industrial extraction of solanesol;NaOH solid insoluble in petroleum ether,its mechanism is not clear.5.Determination of solanesol in potato stems and leaves by different varieties and different harvest Time.The results showed that the content of solanesol was the highest in the growth and development of potato tubers,and the contents of solanesol were the highest in the stems and leaves of the potato.The results showed that the potatoes in the same potato were the same in the same potato planting base in Guyuan,Ningxia.The content of solanesol decreased,and the maximum yield of potato tubers was considered as the main target.The highest content of solanesol in stemss and leaves after ripening of potato tubers was used to reserve the raw materials of solanesolThe innovation work of this paper is as follows:(1)A process for the production of solanesol in potato stems and leaves was established.95% ethanol(80% ethanol available in industrial production)was heated and refluxed twice for 1.5 h each time;(2)The establishment of a broken method of compliance with the test requirements of the potato leaves extract before cellulase enzyme solution that is 50 mg·m L-1 for 3 h;(3)The process of saponification of solanesyl ester in the extract into free solanesol was established,and the related parameters of saponification were optimized and the theoretical basis was provided to improve the content of solanesol;(4)The extraction of solanesol with petroleum ether was carried out to provide theoretical basis for industrial extraction of solanesol;(5)The contents of solanesol in the stems and leaves of different potato varieties were determined,and the data were provided for the extraction of raw materials of solanesol. |
