Objective: To detect the expression of normal protein tyrosine phosphatase PTPN12 protein in non-small cell lung cancer tissues,paracancerous tissues and tissues,and explore the relationship between clinical and pathological features of patients with the expression of PTPN12 protein in non-small cell lung cancer;and combined with clinical follow-up data to have a retrospective analysis of the relationship between PTPN12 expression and patient survival and the recurrence time.Methods: Immunohistochemistry was used to detect the expression of PTPN12 protein in 40 cases of non small cell lung cancer(HCC),and of the adjacent tissues and normal tissues of 40 patients with non small cell lung cancer.SPSS21.0 statistical software package was used to analyze the experimental data,and the relationship between PTPN12 expression and clinicopathological features was analyzed by chi square test and rank correlation analysis.Survival analysis was performed using Kaplan-Meier analysis.Multivariate Cox regression analysis was used to assess the prog-nostic significance of PTPN12 protein expression in patients with non-small cell lung cancer.Results: Compared with normal tissues,the results of non small cell lung cancer were statistically significant(X~2=5.333,P=0.018).Compared with the adjacent tissues,there was no significant diffe-rence between the adjacent tissues and normal tissues(X~2=0.549,P=0.311 ? X~2=2.527,P=0.086).The expression level of PTPN12 and related clinical data for statistical analysis showed that PTPN12 protein expression in tumor tissues of patients was effeted by lymph node metastasis,and in patients with lymph node metastasis the PTPN12 is low,the difference was statistically significant(X~2?=6.337,P=0.012).Compared with the patients with early stage of TNM,theexpression of PTPN12 protein was lower,and the difference was statistically significant.(X~2 ?=0.050,P=0.025).In patients with low tumor differentiation,the expression of PTPN12 protein was lower in those with higher differentiation,and the difference was statistically significant(X~2 ? =0.357,P=0.012).While the PTPN12 protein expression level has no significant correlation with age,gender,pathological type and smoking history.The survival rate and recurrence of 107 patients with NSCLC were statistically analyzed by univariate analysis.The results showed that the overall survival rate of PTPN12 low expression group was lower than that of high one(P=0.035<0.05).The recurrence rate of low expression group was higher than that of high expression group(P=0.015<0.05).Using Cox regression model,the significant variables in the univariate analysis were analyzed,results showed that the low expression of PTPN12 were independent risk factors of survival time of NSCLC patients(HR=4.175,95%CIconfidence interval: 1.034-4.975,P=0.047).At the same time,low expression of PTPN12 is also a high risk factor for recurrence in patients with NSCLC(HR=1.329,95% CI confidence interval: 1.126-3.012,P=0.043).Moreover,Cox regression factor analysis results showed that: tumor differentiation,lymph node metastasis and TNM stage patients are high risk factors affecting the overall survival rate after surgery,and are risk factors for the recurrence of postoperative patients.Conclusion:1 PTPN12 protein is expressed in tumor tissues,adjacent tissues and normal tissues.The expression of PTPN12 protein in non-small cell lung cancer patients was significantly higher than that in the corresponding normal tissues,but there was no significant difference in the expression of adjacent tissues and normal tissues in patients with non-small cell lung cancer.The expression level of PTPN12 protein was correlated with tumor differentiation,clinical stage and lymph node metastasis.These results suggest that the high expression of PTPN12 may inhibit the progression,invasion and metastasis of tumor cells,and PTPN12 may be used as a reference index for predicting theprognosis of patients with non-small cell lung cancer.2 There was no significant difference in the expression of PTPN12 in lung squamous cell carcinoma and the results showed that PTPN12 could not be used as a marker for the prognosis of lung squamous cell carcinoma and adenocarcinoma of lung.3 In non small cell lung cancer,the overall survival rate of PTPN12 high expression group was lower than that of low expression group,and the recurrence rate was low.These results suggest that the low expression of PTPN12 protein is an independent risk factor for postoperative survival and recurrence in patients with non-small cell lung cancer,which confirms that PTPN12 is a new candidate of tumor suppressor gene.The expression of PTPN12 protein in non-small cell lung cancer was not correlated with gender,age,smoking history and other clinical parameters.
