Objective:To detect the expression of normal protein tyrosine phos-phatase PKM2 protein in non-small cell lung cancer tissues,paracancerous tissues and tissues,and explore the relationship between clinical and pathological features of patients with the expression of PKM2 protein in non-small cell lung cancer;and combined with clinical follow-up data to have a retrospective analysis of the relationship between PKM2 expression and patient survival and the recurrence time.Methods:Immunohistochemistry was used to detect the expression of PKM2 protein in 40 cases of non small cell lung cancer(HCC),and of the adjacent tissues and normal tissues of 169 patients with non small cell lung cancer.SPSS22.0 statistical software package was used to analyze the experimental data,and the relationship between PKM2 expression and clinicopathological features was analyzed by chi square test and rank corre-lation analysis.Survival analysis was performed using Kaplan-Meier analysis.Multivariate Cox regression analysis was used to assess the prog-nostic significance of PKM2 protein expression in patients with non-small cell lung cancer.Results:The expression of PKM2 in non small cell lung cancer was statistically significant(X~2=7.627,P=0.031).Compared with normal tissues and adjacent tissues,the results of non small cell lung cancer were statistically significant(X~2=7.366,P=0.007?X~2=0.912,P=0.340).Compared with the adjacent tissues,there was no significant differrence with normal tissues(X~2=3.208,P=0.073).The expression level of PKM2 and related clinical data for statistical analysis showed that PKM2 protein expression in tumor tissues of patients was effected by lymph node metastasis,and in patients with lymph node metastasis the PKM2 is high,the difference was statistically significant(X~2=8.624,P=0.041).Compared with the patients with early stage of TNM,the expression of PKM2 protein was higher,and the difference was statistically significant.(X~2=0.073,P=0.032).In patients with low tumor differentiation,the expression of PKM2 protein was higher in those with lower differentiation,and the difference was statistically significant(X~2=0.571,P=0.019).While the PKM2 protein expression level has no significant correlation with age,gender,pathological type and smoking history.The survival rate and recurrence of 169 patients with NSCLC were statistically analyzed by univariate analysis.The results showed that the overall survival rate of PKM2 low expression group was higher than that of high one(P=0.019<0.05).The recurrence rate of low expression group was lower than that of high expression group(P=0.040<0.05).Using Cox regression model,the significant variables in the univariate analysis were analyzed,results showed that the high expression of PKM2 were independent risk factors of survival time of NSCLC patients(HR=4.175,95%CI:1.034-4.975,P=0.041).At the same time,high expression of PKM2 is also a high risk factor for recurrence in patients with NSCLC(HR=1.329,95%CI:1.126-3.012,P=0.040).Moreover,Cox regression factor analysis results showed that:tumor differentiation,lymph node metastasis and TNM stage patients are high risk factors affecting the overall survival rate after surgery,and are risk factors for the recurrence of postoperative patients.Conclusion:1.PKM2 protein is expressed in tumor tissues,adjacent tissues and normal tissues.The expression of PKM2 protein in non-small cell lung cancer patients was significantly higher than that in the corresponding normal tissues,but there was no significant difference in the expression of adjacent tissues and normal tissues in patients with non-small cell lung cancer.The expression level of PKM2 protein was correlated with tumor differentiation,clinical stage and lymph node metastasis.These results suggest that the high expression of PKM2 may encourage the progression,invasion and metastasis of tumor cells,and PKM2 may be used as a reference index for predicting the prognosis of patients with non-small cell lung cancer.2.There was no significant difference in the expression of PKM2 in lung squamous cell carcinoma and the results showed that PKM2 could not be used as a marker for the prognosis of lung squamous cell carcinoma and adenocarcinoma of lung.3.In none-small cell lung cancer,the overall survival rate of PKM2 high expression group was lower than that of low expression group,and the recurrence rate was high.These results suggest that the high expression of PKM2 protein is an independent risk factor for postoperative survival and recurrence in patients with non-small cell lung cancer,which confirms that PKM2 is a new candidate of tumor proto-onco gene.The expression of PKM2protein in non-small cell lung cancer was not correlated with gender,age,smoking history and other clinical parameters.
