Objective : This study is aimed to detect the expression level of autophagy-related gene Beclinl and anti-apoptotic gene Bcl-2,collected from childhood acute leukemia(AL)patients,and explore their clinical significance in childhood acute leukemia,study the relaionship between the changes of expression level of the two factors and the occurrence and development of childhood acute leukemia,trying to find out the possibility of using Beclinl and Bcl-2 as targets for the treatment of childhood acute leukemia.Methods:To dect the protein level of Beclinl and Bcl-2 expression in BMMNCs,from 30 childhood acute lymphoblastic leukemia?20 childhood acute myelocytic leukemia and 20 non-neoplastic patients,by immunohistochemistry.The result were expressed as positive rate and evaluated with chi-square test,set P<0.05 for the difference is significant.Spearman correlation analysis was performed to evaluate the correlation coefficient between the expression level of the two genesin actue leukemia,p<0.01 was considered as statistical significance.Results:1 The expression positive rate of Beclinl in ALL group,AML group and control group were 66.7%,75.0% and 30.0%,the difference was statistically significant(P <0.05),the positive rate of Beclinl in AML was higer than in ALL,but the difference was no statistically significant(P >0.01),the positive rate of Beclinl in ALL and in AML both higer than in control group,and the difference between the ALL,AML and the control group was statistically significant(P <0.01).2 The expression positive rate of Beclinl in actue leukemia group and in non-neoplastic group were 78.0% and 30.0%,the difference in the two groups was statistically significant(P <0.05).3 The expression positive rate of Bcl-2 in ALL group,AML group and control group were 63.3%,70.0% and 20.0%,the difference was statistically significant(P<0.05),the positive rate of Bcl-2 in AML was higer than in ALL,but the difference was no statistically significant(P>0.01),the positive rate of Bcl-2 in ALL and in AML both higer than in control group,and the difference between the ALL,AML and the control group was statistically significant(P <0.01).4 The expression positive rate of Bcl-2 in actue leukemia and in non-neoplastic patients were 66.0% and 20.0%,the difference in the two groups was statistically significant(P <0.05).5 The expression level of Beclinl and Bcl-2 in acute leukemia was positive correlated,the spearman correlation coefficient was 0.651,P<0.01.Conclusion1 The protein of Beclin1 and Bcl-2 were expressed both in non-neoplastic patients and in childhood acute leukemic patients.The result suggested that autophagy and anti-apoptotic were commonly exist in cells.2 The positive rate of Beclinl was higer in childhood acute leukemic than in non-neoplastic patients,suggesting that the change of the autophagy maybe play a important role,in the occurrence and development of the acute leukemia.3 The positive rate of Bcl-2 was higer in childhood acute leukemic than in non-neoplastic patients,indicating that Bcl-2 was importantly in the process of the acute leukemia.4 The higher positive rate of the Beclinl and Bcl-2 expression in acute leukemic,show that the occurrence and development of the leukemia is regulated by mutiple genes,the autophagy and apoptosis both play a important role in the process.So detect the two genes can contribute to the treatment effect and prognosis of chilodhood acute leukemia.
