CHAPTER ONE : THE EFFECT OF YUXING DILONG DECOCTION ON NICOTINE WITHDRAWALObjective: To observe the effects of Yuxing Dilong decoction on nicotine withdrawal symptoms.Methods:Nicotine addiction model model was successfully established,and animals were divided into following groups randomly: control group?model group and Yuxingdilong decoction high,middle,low does groups.In the nicotine addiction model,the nicotine withdrawal symptoms and the level of dopamine(DA)in the striatum in each group were observed.Results:1?In the nicotine addiction model,compared with the model group,the high dose Yuxingdilong decoction can significantly increase the number of crossing squares,and reduce the times of body erection(P<0.05).2?Compared with the model group,the high,medium and low dose Yuxingdilong decoction can reduce the swimming immobility time,but the difference was not statistically significant(P>0.05).3 ? The DA concentration of five groups were(15.19±3.77)ug/g?(9.01±1.27)ug/g?(10.89±2.49)ug/g?(11.5±2.4)ug/g?(12.78±1.51)ug/g.The difference of DA concentration among the five groups was statistically significant(F=7.017,P=0.000).the middle,high dose Yuxingdilong decoction can significantly reduce the level of DA(P<0.05).Conclusion: Yuxingdilong decoction can improve the withdrawal symptoms,which is hoped to provide new experimental data for the treatment of smoking cessation.CHAPTER TWO : THE PROTECTIVE EFFECT OF YUXING DILONG DECOCTION ON OBSTRUCTIVE PULMONARY DISEASESObjective: To observe the effect of Yuxing Dilong decoction on s moking cession and chronic obstructive pulmonary diseases.Methods:C hronic obstructive pulmonary(COPD)model were successfully establi shed,and animals were divided into following groups randomly: control group?model group and Yuxingdilong decoction high,middle,low d oes groups.The pathological changes and the levels of interleukin 8(I L-8)and tumor necrosis factor alpha(TNF-?)of lung tissue in rats wer e detected.Expression of Cleaved caspase-3 in rat lung tissue were dete cted by immunohistochemistry and western blot.Results:1?In the CO PD model,the alveolus in the COPD rats was damaged obviously and t he infiltration of inflammatory cells was observed.Compared with the model group,low?medium and high dose Yuxingdilong decoction can reduce the mean linear intercept(MLI)(P<0.05).Medium and high dose Yuxingdilong decoction can reduce the destructive index(DI)compar ed with the model group(P<0.05).3?Enzyme-linked immunosorbent as say analysis showed the low?medium and high doses Yuxingdilong dec oction can reduce the levels of IL-8 in lung tissue compared with the model group(P<0.05),the medium and high doses Yuxingdilong decoct ion can reduce the level of TNF-a in lung tissue compared with the mo del group(P<0.05).Immunohistochemistry showed that the percentage of cleaved caspase-3+ cells increased significantly in the model group t han that in control group,the percentage of cleaved caspase-3+ cells in drug treatment group reduced significantly compared with model group(P<0.05);Western blot results showed that the expression of Cleaved caspase-3 protein in lung tissue of rats in model group(0.74 + 0.05)% VS(0.36 + 0.07)% was significantly higher than that of control group(P < 0.05),Cleaved caspase-3 prote of indrug treatment group(low,me dium and high dose group)in the lung tissues of rats(0.69 + 0.04)% a nd(0.44 + 0.06)% and(0.25 + 0.07)% VS(0.74 + 0.05)% were lower than that of model group(P < 0.05).Conclusion: Yuxingdilong decoction can improve the pathological changes of lung tissue,reduce the level of airway inflammation and pulmonary apoptosis..
