Association Of The Macrophage Chemoattractant Protein-1(-2518 G Allele) Gene Polymorphisms With Cancer Risk: A Meta-analysis

Objective: To investigate the relationship between Macrophage Chemoattractant Protein-1(-2518 G Allele)(MCP-1-2518A/G)polymorphism and cancer risk.Methods: We searched Pub Med,Web of science,Embase,VIP,Wan Fang,CBM and CNKI electronic databases,with the last search update on December,2016,to collect all studies on the correlation between-A2518 G in MCP-1gene polymorphism and cancer risk.A total of 30 case-control studies were selected,involving 5602 cases and 6500 controls.All statistical analyses were used by Stata 12.0 software.Q-statistics and I2 statistics were calculated to check the heterogeneity,summary odds ratios(ORs)and 95% confidence intervals(95% CI)were calculated to assess the strength of association.Publication bias was assessed by using Begg' s test and Egger' s test.We also perform the meta-regression and sensitivity analysis.Results: The results indicated that the MCP-1 A2518 G polymorphism had no association with cancer risk.However,stratified analysis by cancer type revealed that MCP-1 gene polymorphism was significantly associated with the risk of hepatocellular cancer,breast cancer,ovarian cancer and NSCLC(P<0.05).Stratified analysis by ethnicity showed that individuals with GG genotypes indicated an increased risk of cancer in Caucasians(GG vs.AG/AA: OR=1.81,95%CI=1.10-2.96),however,there was no association between Asians.This association was also significant in subgroup analyses by source of control,sample size and quality of studies.However,the statistical relationship of polymorphism was not notable in HWE groups and Genotyping method groups.Meta-regression analysis showed that racial differences and the quality of study may be the main sources of heterogeneity among studies.Conclusion: 1.This study suggested that MCP-1-2518 A/G polymorphism may be associated with cancer risk or cancer development in Caucasian.2.MCP-1A-2518 G is expected to be an important molecular marker for early diagnosis and prognosis of cancer.
