Objective:Monocyte chemoattractant protein-1(MCP-1) is a cytokine that exhibits most potent chemotactic activity toward monocytes, which is involved in the recruitment of monocytes into the pathological sites, and induces endothelial cell and monocyte to express the adhesion molecules. Recent studies suggest that MCP-1may accelerate the development and progress of diabetic nephropathy,the activation and infiltration of macrophage induced by over-expression of MCP-1can damage renal tissue through variousmechanisms. Therefore to explore the distribution of monocyte chemoattractant protein-1gene-2518A/G, the single nucleotide polymorphism genotypes and allele frequencies in Guangxi Hans population of China, and to analyze the relationship between the polymorphism of MCP-1-2518A/G and type2diabetic nephropathy, which are likely to become a new clue to the diagnosis and therapy of diabetic nephropathy.Methods:One hundred and thirty patients with type2diabetes mellitus were divided into diabetic nephropathy (DN) group and diabetes without diabetic nephropathy (T2DM) group,and eighty healthy people were selected as normal control (NC) group in Han people of Guangxi province.The genotypes and allele frequencies distribution were assessed by Polymerase chain reaction-restriction fragment length Polymorphism (PCR-RFLP). The values of body height, weight, diastolic blood pressure (DBP), systolic pressure blood (SBP), fasting plasma glucose(FPG), urine albumin excretion rate (UAER), creatinine (Cr), blood urea nitrogen (BUN), high density lipoprotein-cholesterol (HDL-C), total cholesterol(TC), triglyceride(TG), low density lipoprotein-cholesterol (LDL-C) were measured.Results:Three genotypes (AA, AG and GG) of MCP-1-2518polymorphisms were found in Han people of Guangxi province.The frequencies of AA,AG and GG genotypes of MCP-1-2518A/G in healthy controls were38.8%,38.8%and22.4%; whose frenqency of A and G alleles was58.1%,41.9%. The frequencies of AA,AG and GG genotypes of in T2DM were35.3%,39.7%and25.0%;whose frenqency of A and G alleles was55.1%,44.9%. The frequencies of AA,AG and GG genotypes in DN were16.1%,50.0%and33.9%;whose frenqency of A and G alleles was41.1%,58.9%.There was significant difference in the distribution of genetypes and allele frequencies between DN group and NC or T2DM group (P <0.01). The GG genetypes and G allele frequency distributions were higher in T2DM patients with DN than in healthy controls and T2DM patients without DN (P<0.01, P<0.05). The G allele carriers had1.406times risk for increased incidence of DN compared with healthy controls (P<0.01,95%CI1.112~ 1.777). The values of systolic pressure blood (SBP), diastolic blood pressure (DBP),creatinine (Cr), blood urea nitrogen (BUN), urine albumin excretion rate (UAER) were significantly higher in DN group than those of NC or T2DM group (P<0.05). In DN group, the TG, TC and LDL-C levels of AG and GG genotypes patients were significantly higher than the AA genotype patients (P0.05).Conclusions:There is MCP-1gene The polymorphism of-2518A/G single nucleotide polymorphism inChinese Han population of Guangxi province. MCP-1-2518A/G polymorphism is associated with the development of DN in Han population of Guangxi province, and G allele may be a genetic risk factor incidence of DN. MCP-1gene polymorphism may affect the levels of TG, TC and LDL-C, the DN patients who carry G allele are more likely to cause abnormal lipid metabolism...
