Objective To investigate the protective effect of ozone oxidative preconditioning in a rat model of hepatic ischemia reperfusion injury.And to explore whether the protective effect is related to HSP70.Methods24 clean grade Sprague-Dawley Rats(male,8-week-old,250-300 g of body weight)were randomly divided into 4 groups(6 animals per group): sham operation group(group Sham),ischemia/reperfusion group(group I/R),ozone oxidative preconditioning group(group Ozone OP)and quercetin group(group Q).Hepatic ischemia reperfusion injury model was induced by clamping the branches of hepatic artery,portal vein and bile vessel in the median and left lateral hepatic lobes for 45 min,followed by 2hr reperfusion.Group Sham: exposed but not ligation the branches of hepatic artery,portal vein and bile vessel,and closed the abdominal cavity after 45 min.Group Ozone OP: before the I/R procedure,rats received five-day ozone treatments by intraperitoneal injection with ozone and oxygen mixed gas(ozone concentration: 50ug/ml,1mg/kg.d).Group Q:intraperitoneal injection with quercetin(7mg/kg)1hr before ozone oxidative precondioning.After reperfusion,collected blood samples from aorta abdominalis for analysis of serum ALT,AST,IL-1?,IL-6,TNF-?.Executed the rat and hepatic tissue samples were collected for analysis of caspase-3 and apoptotic index.Results Compared with group Sham,concentrations of serum ALT,AST,I L-1?,IL-6and apoptotic index in group I/R,group Ozone OP and group Q were increased(P<0.05),concentrations of serum TNF-? and caspase-3 in group I/R and group Q were increased(P<0.05),concentrations of serum TNF-? and caspase-3were no statistic difference between group sham and group Ozone OP.Compared with group I/R,concentrations of serum ALT,AST,IL-1?,IL-6,TNF-?,apoptotic index and caspase-3 in group Ozone OP were decreased(P<0.05),concentrations of serum ALT,AST,IL-1?,IL-6 and TNF-? in group Q shows no significant difference to group I/R(P>0.05).Compared with group Ozone OP,concentrations of serum ALT,AST,IL-1?,IL-6,TNF-?,apoptotic index and caspase-3 in group Q were increased(P<0.05).Conclusions Ozone oxidative preconditioning could protect the rats against hepatic ischemia/reperfusion injury.The protective effects may related to the increasing activity of HSP70 inhibition the inflammatory response and apoptosis.
