Objective To observe the effects of remifentanil preconditioning on the content of H-FABP in serum and NF-κB,HSP70 in the cardiac muscle tissue of canines with acute myocardial ischemia reperfusion injury(MIRI).To investigate the mechanism of action about remifentanil's anti-MIRI.Methods 24 healthy hybrid canines were randomly divided into four groups(n=6):sham operated group(S),ischemia-reperfusion group(I/R),remifentanil preconditioning groupâ… (RPCâ… ) and remifentanil preconditioning groupâ…¡(RPCâ…¡).The model of MIRI was established by occluding the anterior descending branch of left coronary artery with 30min ischemia and 2h reperfusion.In RPCâ… group,30min's infusion of remifentanil of 1μg·kg-1·min-1 was administrated before ischemia while that was 2μg·kg-1·min-1in RPCâ…¡group.The contents of H-FABP in serum were measured at four time points:Pre-preconditioning,30 min after ischemia,1 h and 2 h after referfusion respectively by enzyme linked immunosorbent assay(ELISA).The expression of NF-κB and HSP70 in the cardiac muscle tissue were measured at 2 h after referfusion by immunohistochemistry.Results Compared with the sham operated group,the contents of H-FABP,NF-κB and HSP70 of the other three groups all heightened significantly(P<0.05).Compared with the schemia-reperfusion group,remifentanil preconditioning groupâ… and remifentanil preconditioning groupâ…¡'levels of H-FABP reduced(P<0.05),and the expression of NF-κB attenuated(P<0.05),but the expression of HSP70 strengthened(P<0.05).Compared with the remifentanil preconditioning groupâ… ,remifentanil preconditioning groupâ…¡'contents of H-FABP reduced(P<0.05),but the expression of NF-κB demonstrated to be no significant variation(P>0.05),however the expression of HSP70 strengthened(P<0.05).Conclusion Remifentanil preconditioning can significantly decrease the levels of H-FABP in serum of canines with acute myocardial ischemia reperfusion injury,can attenuate the activity of NF-κB in the cardiac muscle tissue in the early phase of reperfusion,and can strengthen the expression of HSP70 in the cardiac muscle tissue in the early phase of reperfusion.
