| Objective: The aim of this study was to investigate the association between the polymorphism of N-acetyltransferase 2 gene and the genetic susceptibility of acute leukemia.Methods: Search strategies,strict inclusion and exclusion criteria were made in advance,then we searched Pub Med,EMBASE,Cochrane library,web of science,China biomedical literature database(CBM),China CNKI and Wanfang database before January 1,2017。Extraction and literature screening were conducted independently by two researchers in accordance with strict inclusion and exclusion criteria.The association between N-acetyltransferase 2 gene polymorphisms and genetic susceptibility to acute leukemia was analyzed using Stata12.0 software.Publication bias was examined by the Egger of Stata 12.0 software.Result: 6 articles(1350 case and 4090 controls)were included in this Meta-analysis.The Meta analysis showed significant association between NAT2 acetylation and acute leukemia(slow acetylation OR=1.25,95%CI[1.09,1.44];fast acetylation OR=0.8,95%CI[0.7,0.91]).According to the subgroups of acute leukemia,This analysis showed significant association between NAT2 acetylation and acute lymphoblastic leukemia susceptibility(slow acetylator OR=1.51,95%CI[1.09,2.11];fast acetylation of OR=0.66,95%CI[0.47,0.92]),but not Acute myeloid leukemia(slow acetylator OR=1.23,95%CI[0.86,1.75],fast acetylator OR=0.94,95%CI[0.81,1.09]).In the subgroup by race,association was found in Caucasian(slow acetylator OR=1.51,95%CI[1.04,2.18];fast acetylatior OR=0.66,95%CI[0.46,0.96]),but not found in Asian(Slow acetylatior OR=0.96,95%CI[0.70,1.32];fast acetylatior OR=1.04,95%CI[0.76,1.43]),and nonwhite(slow acetylator OR=1.48,95%CI[0.97,2.26];fast acetylator OR=0.68,95%CI[0.44,1.03]).In the subgroup by age,association was found in child(slow acetylator OR=1.35,95%CI[1.07,1.70];fast acetylator OR=0.74,95%CI[0.56,0.93])and infant(slow acetylator OR=7.91,95%CI[3.87,6.16];fast acetylator OR=0.13,95%CI[0.06,0.26]),but not found in adult(slow acetylator OR=1.1,95%CI[0.92,1.31];fast acetylator OR=0.91,95%CI[0.76,1.09]).Conclusions: The Meta analysis showed that N-acetyltransferase2 gene polymorphisms may be associated with the susceptibility of acute leukemia in Caucasians,NAT2 slow acetylation increased Caucasians suffering from acute lymphoblastic leukemia risk,while the fast acetylation has protective effect.There is age difference in the association between NAT2 acetylation and susceptibility to acute leukemia,NAT2 acetylation is associated with the susceptibility of childhood acute leukemia,and is not associated with susceptibility to adult.There is no evidence that the NAT2 acetylation is associated with susceptibility to acute myeloid leukemia. |
