ObjectiveBy investigating the expression and alteration of the Parkinson disease protein 7(PARK7/DJ-1)in placenta tissue and serum in severe preeclampsia(PE)to find out its association with the pathogenesis of PE,analyze the relationship between PE and serum lipid and discuss the possible roles in the pathogenesis of PE as well as the correlation between the umbilical cord blood levels of DJ-1 and neonatal body weight.MethodsAll 120 cases of pregnant women are admitted into Fujian Provincial Maternity and Child Care Hospital from November 2015 to March 2017,divided into 4groups,30 cases in each group.Severe PE is divided into 2 groups.The firstone is the early-onset group that the gestational week is less than 34 weeks;the second one is late-onset PE group that the incidence of gestational age over 34 weeks.Control group 1: gestational week less than 34 weeks of healthy pregnant women;Control group 2:gestational week over or equal to 34 weeks of no complication of pregnancy with scar uterus,cephalopelvic disproportion,abnormal fetal position of women or social factors for cesarean section.The serum and fetal umbilical cord blood levels of DJ-1 were detected by enzyme-linked immunosorbent assay(ELISA),and the levels of lipid and renal function were detected with biochemistry analyzer.The expression of DJ-1 at mRNA level was detected by real time reverse transcription polymerase chain reaction(RT-PCR),and western-blot was used to examine at protein level.One Way ANOVA,Independent Samples T test or Wilcoxon signed-rank test or Kruskal Wallis Test were used for statistical analysis.Results1.In early-onset and late-onset severe PE groups,the levels of serum total cholesterol(TC),triglyeride(TG),low density lipoprotein cholesterol(LDL)andapolipoprotein B(ApoB)were higher than that of their controls,while high densitylipoprotein cholesterol(HDL)and apolipoprotein AI(ApoA1)were lower than that of their controls(P<0.05).2.The levels of serum DJ-1 was(1.6040±0.465)mmol/L in early-onset PE group and(1.2810±0.419)mmol/L in late-onset PE group,significantly elevated as compared to(1.0698±0.333)mmol/L in early control group and 1.0625±0.329)mmol/L in late control group.3.The levels of umbilical cord blood DJ-1 was(1.3928±1.036)mmol/L in early-onset PE group and(1.4544±0.695)mmol/L in late-onset PE group,significantly elevated as compared to(0.9764±0.390)mmol/L in late control group.4.RT-PCR and western-blot revealed that:the level of DJ-1 expression was significantly higher in early-onset and late-onset preeclampsia groups than control group.5.The serum levels of DJ-1 in early-onset and late-onset severe PE groups were positively correlated with serum TG(P<0.05).6.The cord blood levels of DJ-1 in early-onset and late-onset severe PE groups were negatively correlated with neonatal body weight(P<0.05).Conclusion1.The serum levels of HDL and Apo A1 in preeclampsia group were lower than those in the same control group,while TC,TG,LDL and Apo B water were higher than those in the control group at the same time,suggesting that lipid metabolism disorder is an important clinical manifestation of preeclampsia.2.Dyslipidemia may be associated with increased DJ-1 expression in patients with PE,and may be involved in the pathophysiology of high serum DJ-1 levels led to development of PE.3.The cord blood levels of DJ-1 in early-onset and late-onset severe PE groups were negatively correlated with neonatal body weight,which might affect the intrauterine growth and development of fetus.
