The Association Between Single Nucleotide Polymorphism Of LncRNA And Prognosis In Patients With Primary Hepatocellular Carcinoma

Background and purpose: Long non-coding RNA H19(LncRNA H19)is a well known cancer-related lncRNA.However,the relationships between H19 common genetic variations and survival in patients with primary hepatocellular carcinoma(HCC)remain poorly understood.In this study,we investigated the relationship between single nucleotide polymorphism(SNP)rs217727 in H19 gene and survival in resected primary HCC.Methods: This study contained 399 tissue samples from resected primary HCC patients,and all of the samples were subjected to do the genotype for rs217727 in H19 gene.The data were then analyzed and the relationships between individual genotypes and survival in this group of patients were examined by statistical tests.Results: CC,CT and TT genotypes composed the SNP rs217727.151 out of 399 case(37.84%)were CC carriers,201 out of 399 cases(50.38%)were CT carriers,and 47 out of 399 cases(11.78%)were TT carriers.The genotypes of rs217727 showed correlation with patient survival rate(alive vs.dead,p<0.001),but no correlation with with patients' age and gender(?55 vs.>55,p=0.804;male vs.female,p=0.967)as well as no correlation with tumor pathological characteristics including TNM stage(I/II vs.IIIa,p=0.134),tumor sites(left vs.light,p=0.611),tumor encapsulation(absent vs.present,p=0.376),vascular invasion(absent vs.present,p=0.051),tumour size(diameter)(?5,5-10 and >10cm,p=0.096),and serum AFP level(?400 vs.>400(ng/m L,p=0.255)(Table1)examined by Chi-square test,suggesting that SNP rs217727 of H19 may be correlated with survival in patients with primary HCC.The TT genotype of rs217727 was significantly related with survival.This group of patients had a total of 42.86% overall death rate;whereas,patients carrying TT genotype only had a 19.15% overall death rate which decreased 23.71%(42.86% vs.19.15%,p=0.002).Further analysis showed that patients carrying TT genotype had significantly decreased death risk with an adjusted hazard ratio(HR)0.21,95% CI(0.106-0.445)which was significantly lower than patients with CT genotype had(P=0.000).Furthermore,survival analysis by Kaplan–Meier curve plus the log-rank test showed that patients carry TT genotype had significantly longer over survival time than CT and CC carriers(P=0.019).Conclusions: The rs217727 SNP located in H19 is closely related to clinical prognosis of patients with primary HCC.The rs217727 SNP located in H19 may be used as an novel prognostic biomarker for assessing survival in patients with.resected primary HCC.
