Background: Astrocytic tumours are the most common types of tumours of neuroepithelial tissue,According to histopathological and molecular characteristics,astrocytic tumours are classified into four grades(WHO I–IV)by World Health Organization(WHO),the mortality rate increases with the increase of tumor grade,and the patients' survival shortened.Therefore astrocytic tumours become a tumor with the high mortality rate in the intracranial tumor.Recently incidence rate of astrocytic tumours are increasing year by year,which threat seriously to human health.According to the different of tumours' grade,the treatment is different as well,however,the astrocytic tumours treatment preferred surgical resection.Because of the diffusely to the surrounding infiltration,located in the cerebrum function area and tumor located in the hardly reach part of cerebrum,apart from the pilocytic astrocytoma(WHO I),satisfactory effect can be got only by surgery,synthetic therapy of surgery combined withchemotherapy and radiotherapy should be employed to enhance the other grade astrocytic tumours' survival rate.As well known,high-grade astrocytomas,especially glioblastoma,due to the serious invasion of brain tissue around,and no clear dividing line with brain and nerve tissue,it will still have a considerable amount of residual tumor,so looking for a more effective target is the key to the treatment of astrocytic tumours.VEGF is an important index in the study of central nervous system tumors,which is of great significance to the diagnosis and treatment of tumors.The expression levels of D2-40 and STAT6 were closely related to the occurrence and progression of tumor,the degree of malignancy,the degree of invasion,and the sensitivity to radiotherapy and chemotherapy.Recently,the relationship between expression of this two indexes and prognosis and treatment of lung cancer,colon cancer and liver cancer was researched on,but the study of astrocytic tumours was rare.Objective: Detected the expression of VEGF,D2-40 and STAT6 in astrocytic tumours by immunohistochemical staining,to study the relationship between the expression of VEGF,D2-40 and STAT6 and histologic grade in patients of astrocytic tumours,and to explore the relationship between VEGF,D2-40,STAT6 and development,progression,invasion of astrocytic tumours.Methods: Paraffin embedded specimens of 120 cases astrocytic tumours patients were collected from the Tianjin Huanhu hospital neurosurgery in 2012-2014,including pilocytic astrocytoma(30 cases),diffuse astrocytoma(30 cases),anaplastic astrocytoma(30 cases)and glioblastoma(30 cases).All specimens had initially been selected for construction of tissue microarrays(TMA)for use in future research.Immunohistochemical method was used to detect the expression of VEGF,D2-40 and STAT6 in 120 cases astrocytic tumours patients,analyzed the datas by statistical software SPSS 17.0 to explore the expression and clinical significance of them in astrocytic tumours.Results: 1.There was a significant difference in the positive of VEGF and STAT6 between high grade astrocytomas(histological grading ?~?)and low grade astrocytomas(histological grading ?~?),P<0.05.The positive reaction rate of VEGF and STAT6 was increased with the increase of tumor malignancy.2.The expression of D2-40 in astrocytic tumours was not committed to the lineage,however there was a significant difference between glioblastoma(histological grading ?)and anaplastic astrocytoma(histological grading ?),P<0.05.3.The expression of VEGF was positively correlated with the expression of D2-40 and STAT6 in astrocytic tumours,which had significant statistical significance(r=0.645,P=0.000 vs r=0.637,P=0.000);The expression of D2-40 was positively correlated with the expression of STAT6 in astrocytic tumours,which had significant statistical significance(r=0.611,P=0.000).Conclusion: The expression of D2-40 and STAT6 play a very important role in development and growth of astrocytic tumors.And the expression of VEGF reflect the general rule of tumor angiogenesis and cell proliferation.The expression of VEGF was positively correlated with the expression of D2-40 and STAT6,and the expression of D2-40 was positively correlated with the expression of STAT6.Combined detection of this three indexes can evaluate the biological behavior and prognosis of the patients more exactly,and provide the basic theory for the targets treatment of astrocytic tumours.
