Aims: Analyzing ABO subgroups' results of serological and sequencing test of blood donors,in order to obtain the serological characteristics and allele mutation sites of ABO subgroups in blood donors of Binhai district.Methods: Using routine ABO blood typing test to screen the difficult samples;then using classic serological tube method to indentified those difficult samples.Sending the real ABO subgroup samples to Sangon Biotech(Shanghai)Co.,Ltd.to carry on the molecular biological tests.After sequencing,those who had missense mutation then carried on the protein function analysis,and using PyMOL software to setup Spatial model of glycosyltransferases.Results: A total of 45880 blood donors' ABO blood type were routinely tested.From them 20 phenotypes were indentified to be ABO subgroup by standard serologic techniques: A2(1)?AX(2)?Bx(3)?Bm(1)?B3(1)?A2B(2)?AXB(4)?ABx(3)?B(A)(2)and cisAB(1).By DNA analysis and sequencing,among 20 variants 10 genotypes were dected with nucleotide mutation(1009A>G?905 A>G?904 A>G?503G>T?28G>A?588C>G?550G>A?700C>G?640A>G?803G>C)which led to animo acid substitution: R337G?D302G?K314E?R168W?G10R?C196W?V184M?P234A?M214V?G268A.The amino acid substitutions caused by missense mutations are conserved amino acids in evolution and the spatial structure of the side chain of amino acids were changed greatly after the amino acid replacement.Conclusions:(1)ABO subgroups in serological detection often appears the antigen and(or)antibody's abnormal reduce and(or)increase.So we need to know the serological characteristics of ABO subgroups in order to make rapid and accurate identification.The mutation of ABO alleles caused amino acid substitutions of glycosyltransferases A and glycosyltransferases B.(2)After amino acid's replacement,the change of amino acid's physical and chemical properties,hydrophobic and charge properties,intermolecular force between amino acid side chain,had effects on GTA's and GTB's catalytic activity and specificity,thus forming those ABO subgroups.
