Experimental Study Of Effect Of Shentong Zhuyu Decoction On The Proliferation Of Synovial Fibroblasts And Expression Of COX-2 Protein

Objective: To observe the effects of Shentong Zhuyu decoction on the proliferation of osteoarthritis(OA)synovial fibroblasts and expression of cyclooxygenase-2(COX-2)protein.Methods: Synovial fibroblasts were separated and identified by flow cytometry.Synovial fibroblasts were cultured in saline(negative control),etoricoxib containing serum(positive control)and Shentong Zhuyu decoction contained serum.The proliferation ability of synovial fibroblasts were detected by MTT.The expression of COX-2 protein was detected by Western-blot.Results: Compared with negative control,etoricoxib containing serum and Shentong Zhuyu decoction contained serum both can inhibit the proliferation ability of synovial fibroblasts(P<0.05),and there has no significance difference between etoricoxib containing serum and Shentong Zhuyu decoction contained serum(P>0.05).Western-blot shows that etoricoxib containing serum and Shentong Zhuyu decoction contained serum both can inhibit the expression of COX-2 protein,and there has no significance difference between etoricoxib containing serum and Shentong Zhuyu decoction contained serum(P>0.05).Conclusion: Shentong Zhuyu decoction contained serum can inhibit the proliferation ability of synovial fibroblasts and the expression of COX-2 protein,which may be one of the potential mechanisms for the treatment of OA.
