Objective: To detect the Borna Disease Virus(BDV)infection condition in brain glioma patients in Zunyi city of Guizhou province,then to study the clinical characteristics of the brain glioma patients whose BDV P40 gene is positive.Methods: The cerebral glioma tissues and peripheral blood lymphocyte(PBL)from 22 brain glioma patients',24 cases of normal brain tissues,and 24 cases of PBL from healthy people's were collected from the Affiliated Hospital of Zunyi Medical University,during October 2015 to September 2016.The P40 gene fragment of BDV-RNA was from the cerebral glioma tissues and PBL of the brain glioma patients',which was detected by real-time reverse transcription polymerase chain(RT-PCR).And the BDV P40 positive samples were purified,cloned and sequenced,then compared with the foreign standard strains and epidemic strains,analyzed the homology and variation.Results: It was found that the positive rate of the BDV P40 gene fragment in the 22 cases of cerebral glioma tissues was 18.2%,and it was higher than that of the normal brain tissues'(0%).The positive rate of the BDV P40 gene fragment in the PBL of brain glioma patients was 9.1%.The sequencing results of the BDV p40 positive samples from brain glioma and PBL were imported into the National Central of Biotechnology Information(NCBI)system to analyze their homologies with the foreign standard strains and epidemic strains.The BDV P40 gene fragments(nt242-320)compared with Strain V exhibited 97.2% homology,and 3 genetic loci with consistently silent mutations were demonstrated(nt262G>C,nt303 C>T and nt313 A>G),the mutation rate was 3.8%.When it compared with He/80 exhibited 100% homology,2 genetic loci with consistently silent mutations were demonstrated(262 ntG> C,nt313A>G),the mutation rate was 2.5%.However,no changes in the encoded amino acid were noted.The clinical characteristics of brain glioma patients with the BDV P40 positive was the same as the negative one‘s.Conclusion: BDV infection really exists in brain glioma patients in Zunyi city,and the infection of BDV may relate to the brain glioma.The BDV P40 gene fragment is very conservative and rare mutational,it may be a specific marker to prove the patients who really infected by BDV.The clinical characteristics of glioma patients with BDV positive is the same as the patients with BDV negative.
